F-β能够抑制T细胞免疫应答,导致巨噬细胞去活化和组织损伤,所以有人认为TGF-β的抑制剂可以作为结核病化疗的免疫佐剂。两种TGF-β天然抑制剂得克因(decorin)、灭活相关肽(LAP)具有潜在调变TGF-β的作用[17]。得克因是一种小相对分子质量的糖原,可以与TGF-β的生物活性中心结合,阻断TGF-β的活性。在肾小球肾炎的大鼠模型中,得克因的应用可以降低尿蛋白,显著地改善组织病变。LAP是TGF-β的灭活相关肽,它具有结合和灭活TGF-β的能力。重组LAP已经应用于能够过度表达TGF-β的转基因鼠,实验结果提示LAP能够阻断TGF-β对肝细胞的增殖效应。已有实验证明得克因和LAP应用于结核病患者可以恢复T细胞的免疫应答能力,改善单核巨噬细胞的免疫效应功能。因此,得克因和LAP可望作为免疫治疗佐剂应用于结核病患者的临床治疗,尤其是对难治性结核病患者特别适用。 作者单位:沈永泉(261042山东省潍坊医学院免疫学教研室)
