【摘要】 目的探讨夜香树不同提取物对小鼠白血病细胞株L1210细胞的增殖抑制作用及机制。方法采用四甲基偶氮唑盐比色(MTT)法、平板集落法和Hoechst33258荧光染色法观察夜香树提取物对L1210细胞的生长抑制和凋亡的影响。结果夜香树提取物中正丁醇提取物和多糖提取物对L1210细胞有浓度依赖性的细胞毒性作用,在终浓度为62.5 μg·ml-1范围,其正丁醇提取物和多糖提取物对L1210细胞的抑制率大于90%,IC50均小于10 μg·ml-1,夜香树正丁醇和多糖提取物能抑制L1210细胞集落形成,并可诱导L1210细胞发生凋亡。结论夜香树提取物对小鼠白血病细胞株L1210细胞的增殖抑制可能通过诱导L1210细胞发生凋亡而发挥作用。

Inhibitory Effect of Extracts from Cestrum nocturnum Linn. on Proliferation of Leukemic L1210 Cells

Abstract:ObjectiveTo evaluate the inhibitory effect of different extracts from Cestrum nocturnum Linn..(CN) on mouse leukcmic L1210 cells and explore its mechanism. MethodsThe MTT assay, colony forming experiment and Hoechst 33258 fluorescence measurement were used to analyze the effect of extracts from CN on leukcmic L1210 cells. ResultsThe n-bulty alcohol part and polysaccharide part extracted from CN had dose-dependent effects of cytotoxicity.When the concentration was 62.5 μg·ml-1, the inhibitory rate of cells growth was above 90% and the IC50 was below 10 μg·ml-1.The inhibitory rate of cells had remarkable dose-dependence. They both could inhibit colony information and induce apoptosis of L1210 cells. ConclusionThe inhibitory effect of extracts from CN.on leukemic L1210 cells prolifertion may be related with apoptosis.
