一氧化氮合酶抑制剂对大鼠异氟醚MAC的影响 |

【摘要】 目的 研究神经元型一氧化氮合酶(nNOS)抑制剂7-硝基吲唑(7-NI)对大鼠异氟醚MAC的影响。方法 30只雄性SD大鼠,随机分为6组(每组5只)。大鼠置于密闭麻醉系统中,按标准夹尾法测定异氟醚MAC。每只鼠测定基础MAC后,随机经腹腔注射3ml花生油,或20、60、80、120或250mg.kg-1 7-NI(溶于3ml花生油),30分钟后测定MAC值。选120mg.kg-1 7-NI组大鼠再腹腔注射500mg.kg-1L-精氨酸,15分钟后测定MAC值。结果 20~250mg.kg-1 7-NI显著降低异氟醚MAC,分别较基础MAC下降(15.6±2.3)%、(21.9±2.1)%、(29.9±2.5)%、(30.4±1.7)%、(29.8±3.3)%,当7-NI剂量超过80mg.kg-1时出现封顶效应。500mg.kg-1 L-精氨酸可完全逆转120mg.kg-1 7-NI降低MAC效应。结论 抑制中枢nNOS活性,可增强异氟醚麻醉效能,提示中枢NO信息途径可能在全麻药作用分子机制中起作用。 【关键词】 异氟烷 投药和剂量 一氧化氮合酶 吲唑类 药理学
Effects of nitric oxide synthase inhibitor 7-nitro indazole on the threshold for isoflurane anesthesia in rats

【Abstract】 Objective To investigate the effects of neuronally selective nitric oxide synthase(NOS)inhibitor,7-nitro indazole(7-NI),on the minimum alveolar concentration (MAC) of isoflurane anesthesia.Methods Thirty Sprague-Dawley rats were divided randomly into six groups.The test was conducted in a closed anesthetic system.The MAC was determined by observing the response to tail clamp.Arachis oil(3ml) or 7-NI(20-250mg.kg-1,dissolved in 3ml arachis oil) were administered intraperitoneally after determining baseline of MAC,30 min later MAC was determined in the presence of NOS inhibitor.Results 20,60,80,120,250 mg.kg-1 7-NI caused a dose-dependent decrease in isoflurane MAC by (15.6±2.3)%,(21.9±2.1)%,(29.9±2.5)%,(30.4±1.7)% and (29.8±3.3)%,respectively,as compared with the baselines.The peak effect was observed at 7-NI dose greater than 80 mg.kg-1.Conclusion Inhibition of the central NOS activity decreases the MAC of isoflurane,indicating that the NO signal pathway plays a role in mechanisms of anesthesia and analgesia. 【Key words】 Isoflurane Administration and dosage Nitric-oxide synthase Indazole Phar[1] [2] 下一页 上一个医学论文: 咪唑安定及异丙酚对大鼠心肌细胞电生理活动的影响 下一个医学论文: 兔关节软骨细胞转化生长因子-β1免疫组织化学的研究
