严重烧伤早期中性粒细胞在肝脏中的聚集机制及参与肝脏损害的研究 |

【摘要】 目的 研究中性粒细胞在肝脏中的聚集、滞留机制及参与肝细胞损伤的作用。方法 应用生物力学技术、流式细胞技术等方法,观察了烧伤病人血清对培养人肝细胞和肝窦内皮细胞间粘附力,以及P-选择素及细胞间粘附分子-1(ICAM-1)在两种细胞中表达情况的影响。结果 (1)严重烧伤后早期中性粒细胞在肝脏中有聚集;(2)肝细胞和中性粒细胞在烧伤血清作用下粘附力以2小时为最强;(3)肝细胞正常情况下无P-选择素和ICAM-1表达,烧伤血清作用后ICAM-1表达增强。肝窦内皮细胞在正常情况下有少量P-选择素和ICAM-1表达,烧伤血清作用后表达增加。结论 严重烧伤后中性粒细胞在肝脏中的渗出和聚集及加重肝脏功能损害,是中性粒细胞、细胞因子和粘附分子协同作用的结果。 【关键词】 烧伤 中性粒细胞 粘附力 P-选择素 细胞间粘附分子-1
Study on Mechanism of Neutrophil Aggregation in Liver and Its Effect on Hepatocytic Injury WANG Xiao-jun, LUO Xiang-dong, LUO Qin, et al. Burn Research Institute, Southwestern Hospital, Third Military Medical University, Chongqing 400038 【Abstract】 Aim To study the mechanism of neutrophil aggregation in the liver and its effect on hepatocytic injury after severe burns during early postburn phase. Methods The change of intercellular adhesive force between neutrophils and hepatocytes, and the changes of the expression of P-selectin and intercellular adhesion molecule-1 (ICAM-1) on the hepatocytes and hepatic sinusoidal endothelial cells (HSECs) were observed dynamically with flow cytometry and micropipette aspiration system under the influence of the burn patient s serum. Results (1)There were polymorphonuclear neutrophils (PMNs) in the liver at the very early stage after burn. (2) In the presence of burn patient s serum, the intercellular adhesive force between neutrophils and hepatocytes reached its top level at 2 hour after co-cultivation. (3) There were no P-selectin or ICAM-1 expression in the normal hepatocytes. Under the influence of burn patient s serum, the expression of P-selectin increased significantly in HSECs but not in hepatocytes,and the expression of ICAM-1 increased significantly in hepatocytes and HSECs.
