冷冻同种异体肌腱小鼠移植的细胞毒 IL 2水平及组织学观察 |

【摘要】 目的 了解冷冻同种异体肌腱移植术后宿主的免疫排斥反应。方法 采用BALB/C小鼠肌袋模型,植入冷冻C57小鼠跟腱,观察术后各期宿主血清微量细胞毒性、局部组织浆IL-2水平和局部组织学变化。结果 (1)经冷冻/冻干处理组淋巴细胞毒性、IL-2活性显著低于未处理组(P<0.01),但仍高于自体组(P<0.01)。(2)处理各组间细胞毒性无差异(P>0.05),而IL-2活性-196℃组高于其他组(P<0.01)。(3)各组局部组织淋巴细胞浸润程度与细胞毒反应相符。结论 经冷冻/冻干处理后,同种异体肌腱移植免疫排斥反应显著降低,但未消失。 【关键词】 移植,同种 移植免疫学 腱 细胞毒性,免疫 白细胞介素-2 组织学
An Investigation of Cytotoxicity, IL-2 and Histology Following Frozen Allografts Tendons in Mice

【Abstract】 Objective In order to investigate the immune rejection response of host to the implanted achilles tendon allografts of C57 mice in BALB/C mice model. Methods The dryly frozen, deeply frozen (-196℃) and frozen (-80℃) achilles tendons of C57 mice were respectively treated as Group A,B,C while fresh tendons allografts served as positive control (Group D) and autografts as negative control (Group E). Immunologic and histologic characters were observed at 2, 7, 14, 28, 42 days, separately. Results (1) The contents of lymphocytotoxicity and IL-2 showed markedly lower in Group A,B,C than in Group D (P<0.01), but were still higher than in Group E. (2) The IL-2 level was significant difference (P<0.01) between Group B and A or C. (3) The histologic immuneresponse in Group A,B,C abated more greatly than in Group D, but the local tissue was still infiltrated by monocytes and lymphocytes. Conclusions The immune rejection response is abated markedly but does not disappear in the treated achilles tendons of allografts. 【Key words】 Transplantation, homologous Transplantation Immunology Tendon Cytotoxicity, immunologic Interleukin-2 Histology
同种异体肌腱移植的临床应用日益受到重视,但对其植入后的免疫反应报道不一。笔者进行了如下实验,以观察经冷冻/冻干处理的同种异体小鼠跟腱在宿主体内的免疫排斥反应。
