【摘要】 目的 探讨褪黑素(melatonin,MT)对重症急性胰腺炎(severe acute pancreatitis,SAP)大鼠胰腺组织中 Smac/DIABLO、XIAP mRNA表达的影响及可能机制。方法 54只SD大鼠随机分成假手术(SO)组、重症急性胰腺炎(SAP)组以及MT干预SAP(MS)组,每组18只。通过胰胆管逆行注射3.5%脱氧胆酸钠方法制备大鼠SAP模型,MS组制造模型前30 min腹腔注射MT。收集各模型组3 h、6 h、12 h标本,观察胰腺组织,检测胰腺组织细胞凋亡,实时定量(real-time)PCR分析胰腺组织中Smac/DIABLO、XIAP mRNA的表达。结果 SAP组和MS组胰腺组织损伤较SO组严重。与SAP组相比,在3 h、6 h、12 h各时点MS组胰腺病理损伤明显减轻,细胞凋亡指数下降(P<0.05),胰腺组织中Smac/DIABLO mRNA表达下调(P<0.05),XIAP mRNA表达上调(P<0.05)。结论 MT能明显改善SAP胰腺组织损伤,对急性胰腺炎有保护作用;这可能与调节Smac/DIABLO、XIAP mRNA的表达有关。

Experimental investigation on the protective effect of MT in severe acute pancreatitis in rats

Abstract Objective To investigate the effect of MT on the mRNA expression of Smac/DIABLO, XIAP in severe acute pancreatitis and its possible mechanism in rats. Methods Fifty-four SD rats were randomly divided into three groups: sham-operation (SO) group, SAP group and MT-pretreated (MS) group. SAP group were induced by retrograde injection of 3.5% sodium deoxycholate into the pancreatic duct. MS group were given intraperitoneal injection with MT 30 min before the induction of SAP. The specimens at 3 h, 6 h, 12 h were collected, the pathological changes of the pancreas were observed, the apoptosis of pancreas were detected and the mRNA expression of Smac/DIABLO and XIAP were analyzed by real-time PCR in pancreas. Results Compared with the SO group, the pancreas pathological changes in SAP group and MS group were significantly aggravated. Compared with SAP group, the pathology injury of pancreas in MS group was obviously alleviated, apoptotic index were markedly decreased (P<0.05), the mRNA expression of Smac/DIABLO were greatly down-regulated (P<0.05) and the mRNA expression of XIAP were sig
