【摘要】 目的 探讨经典神经递质去甲肾上腺素(norepinephrine, NE)对人胰腺癌细胞株MiaPaCa2侵袭能力的影响及其作用机制。方法 取人胰腺癌细胞株MiaPaCa2为研究对象,实验分为空白对照组、0.1μmol/L NE干预组、1μmol/L NE干预组和10μmol/L NE干预组。干预48h后,采用Transwell侵袭实验检测各组中MiaPaCa2细胞侵袭能力的变化;RTPCR法检测细胞中MMP2、MMP9和VEGF mRNA的表达;免疫细胞化学法检测细胞中MMP2、MMP9和VEGF蛋白的表达。结果 1μmol/L NE干预组和10μmol/L NE干预组MiaPaCa2细胞的侵袭能力均高于空白对照组(P<0.01);1μmol/L NE干预组和10μmol/L NE干预组MiaPaCa2细胞中MMP2、MMP9和VEGF mRNA和蛋白表达水平均高于空白对照组(P<0.05)。结论 NE可以通过上调MMP2、MMP9和VEGF的表达从而促进胰腺癌细胞发生远处侵袭转移。

Effect of norepinephrine on the invasiveness of

Methods The pancreatic cancer cell line MiaPaCa2 was used in the research. The cells were randomly divided into control group, 0.1μmol/L NE intervention group, 1μmol/L NE intervention group and 10μmol/L NE intervention group. After intervention for 48h, Transwell invasiveness test was given to examine the changes of invasive ability of MiaPaCa2. The expressions of MMP2 mRNA, MMP9 mRNA and VEGF mRNA were measured by semiquantitative RTPCR; the levels of MMP2, MMP9 and VEGF proteins were measured by immunocytochemical method. Results The invasive ability of MiaPaCa2 in 1μmol/L NE intervention group and 10μmol/L NE intervention group was significantly higher than that in control group (P<0.01). The levels of MMP2, MMP9 and VEGF mRNA and protein expression of MiaPaCa2 in 1μmol/L NE intervention group and 10μmol/L NE intervention group were higher than those in control group (P<0.05).
