【摘要】 目的 比较树突状细胞(DCs)诱导的细胞毒性T淋巴细胞(CTLs)、细胞因子诱导的杀伤细胞(CIK细胞)和淋巴细胞因子激活的杀伤细胞(LAK细胞)对神经胶质瘤的体外杀伤作用。方法 向外周血单个核细胞分别加入不同细胞因子组合,诱导出DCs、CIK细胞和LAK细胞,DCs负载胶质瘤抗原后激活CTLs,MTT法检测对胶质瘤细胞的体外杀伤效应。结果 CTLs对胶质瘤细胞的杀伤作用明显强于CIK细胞和LAK细胞(F=6.42~8.26,q=8.02~10.18,P<0.01);CIK细胞对胶质瘤的杀伤作用明显强于LAK细胞(q=8.43~9.24,P<0.01)。CIK细胞对K562细胞的杀伤作用明显强于LAK细胞和CTLs(F=5.36~7.69,q=7.23~9.56,P<0.01),LAK细胞对K562细胞的杀伤作用明显强于CTLs(q=8.83~9.15,P<0.01)。结论 从人外周血中诱导出的DCs负载胶质瘤抗原后,激活的CTLs在体外对胶质瘤细胞能产生高效而特异的杀伤作用,明显强于CIK细胞和LAK细胞,为临床应用DCs瘤苗奠定基础。

CONTRAST AMONG ANTITUMOR EFFICACY OF LAK, CIK AND CTLs ACTIVATED BY DENDRITIC CELLS GONG ANJING, MENG QINGHAI, ZHU XIANGHUA(Department of Neurosurgery, The Affiliated Hospital of Qingdao University Medical College, Qingdao 266003, China) [ABSTRACT]ObjectiveTo contrast the antitumor efficacy among LAK, CIK and CTLs activated by dendritic cells in vitro. MethodsPeripheral blood monocytes were induced to DCs, CIK and LAK by different kinds of cytokine. After being pulsed in vitro with glioma lysates, DCs were used to induce T lymphocytes into cytotoxic T lymphocytes for glioma cells. The cytotoxicity was assayed by MTT.ResultsCTLs were stronger than CIKs and LAKs in killing glioma cells (F=6.42-8.26,q=8.02-10.18,P<0.01). CIK was stronger than LAK, as well as LAK and CTLs in killing K562 and glioma cells, respectively (q=8.43-9.24,P<0.01) (F=5.36-7.69,q=7.23-9.56,P<0.01). LAK killed more K562 cells than CTLs did (q=8.83-9.15,P<0.01).ConclusionThe antigen specific CTLs can kill glioma cells effectively and specifically ,which might play a great role in clinical therapy.

胶质瘤是中枢神经系统内最常见的恶性肿瘤,由于呈浸润性生长,手术难以切除干净,复发率高,化疗、放疗等其他常规治疗效果都不甚理想。近来发现,免疫治疗对胶质瘤有较好的疗效。目前研究较多的有淋巴细胞因子激活的杀伤细胞(LAK细胞)、细胞因子诱导的杀伤细胞(CIK细胞)及树突状细胞(DCs)诱导的细胞毒性T淋巴细胞(CTLs)。本文比较了上述三种杀伤细胞对神经胶质瘤的体外杀伤作用,并对其可能的机制进行了探讨。
