【摘要】 目的:观察β1,4-GalT I mRNA在坐骨神经损伤后相应节段脊髓和背根神经节中的表达变化。方法:采用real-time PCR 方法, 分析β1,4-GalT I mRNA在大鼠坐骨神经损伤后相应脊髓和背根神经节中的表达变化。运用原位杂交检测β1,4-GalT I在脊髓和背根神经节中的定位情况。结果: Real-time PCR 显示,与正常脊髓和背根神经节相比, β1,4-GalT I在坐骨神经损伤后发生表达变化。原位杂交结果显示β1,4-GalT I在正常脊髓中的表达较弱,主要表达于脊髓灰质中,而损伤后在灰质和白质中均可见有大量的阳性信号。β1,4-GalT I在背根神经节中则主要定位于神经元。结论: 本实验发现β1,4-GalT I在正常脊髓和背根神经节中有表达, 并在坐骨神经损伤后发生表达变化。提示β1,4-GalT I在周围神经损伤后再生和退变过程中可能发挥一定的作用。

Expression of β1,4-GalT I in rat spinal cord and dorsal root ganglion after sciatic nerve injury

[Abstract] Objective: To observe the expressive changes of β1,4-GalT I mRNA in corresponding spinal cord and dorsal root ganglion after sciatic nerve injury. Methods: Real-time PCR was used to detect the expressive changes of β1,4-GalT I mRNA in corresponding spinal cord and dorsal root ganglion after sciatic nerve injury. The localization of β1,4-GalT I mRNA in corresponding spinal cord and dorsal root ganglion was explored by using in situ hybridization. Results: Real-time PCR revealed that, compared with the normal control group, the expressive levels varied after sciatic nerve injury. In situ hybridization showed that the expression of β1,4-GalT I in normal spinal cord was weak and mainly located in the grey matter of spinal cord, and the expression signals were enhanced in spinal cord after sciatic nerve injury and distributed in the grey and the white matter. β1,4-GalT I mainly located in neurons in dorsal root ganglion. Conclusion: β1,4-GalT I is present in normal spinal cord and dorsal root ganglion, and it varies after sciatic nerve injury.
