【摘要】 目的 研究ING4及HIF1α在星形细胞瘤中的表达,探讨它们在星形细胞瘤发生和发展中的作用。方法 采用免疫组化法检测45例星形细胞瘤标本及11例正常脑组织中ING4和HIF1α的表达。结果 ING4在正常脑组织和星形细胞瘤中的阳性表达率分别为100%、42.4%;Ⅰ~Ⅱ级和Ⅲ~Ⅳ级星形细胞瘤中的阳性表达率分别为58.3%、23.8%,差异均有统计学意义(P<0.05)。HIF1α在正常脑组织和星形细胞瘤中的阳性表达率分别为0、71.1%;在Ⅰ~Ⅱ级和Ⅲ~Ⅳ级星形细胞瘤中的阳性表达率分别为54.2%、90.5%,差异均有统计学意义(P<0.05)。结论 ING4和HIF1α基因的表达与星形细胞瘤分级有密切关系,在其发生和发展过程中起重要作用。

【Abstract】 Objective To research the expressions of ING4 and HIF1α in astrocytoma, and explore their roles in genesis and development of astrocytoma. Methods Immunohistochemistry was used to detect the expressions of ING4 and HIF1α in 45 cases of astrocytoma and 11 cases of normal brain tissues . Results The positive rates of the expression of ING4 in normal brain tissue and astrocytoma were 100%,42.4% respectively. The positive rates of the expression of ING4 in gradeⅠ~Ⅱand Ⅲ~Ⅳgrade astrocytoma were 58.3%, 23.8% respectively, the difference was significant(P<0.05).The positive rates of the expression of HIF1α in normal brain tissue and astrocytoma were 0,71.1% respectively. ING4基因是2003年发现的一种强的候选抑癌基因[1],它通过抑制肿瘤血管增生、调控细胞增殖和凋亡及细胞的接触抑制等抑制肿瘤细胞的生长。缺氧诱导因子1α(HIF1α)是一种重要的转录因子,能促进肿瘤的血管生成,对肿瘤迅速增长起着重要的作用。本实验通过对45例星形细胞瘤标本中ING4和HIF1α蛋白检测,研究二者在星形细胞瘤发生和发展过程中的作用,为进一步揭示肿瘤的发病机制、诊断及治疗提供一定的理论依据。
