【摘要】 目的: 探讨细胞周期蛋白Skp2表达与膀胱移形细胞癌生物学行为的关系。方法: 对73例膀胱移形细胞癌进行组织学分级及分期,另取22例正常膀胱组织作为对照,采用免疫组织化学方法SP法检测各例组织中Skp2蛋白表达水平,并分析其表达水平与临床病理特征的关系。结果: Skp2蛋白阳性表达率为50.7%(37/73), Skp2蛋白表达与肿瘤组织学分级、病理分期显著相关。结论: Skp2是评估膀胱移形细胞癌恶性程度,估计预后有价值的指标。

【ABSTRACT】 Objective: To investigate the significance of the protein expression of Skp2 in the progression and prognosis of bladder transitional cell carcinoma.Methods: The pathological stage and histological grade were assessed in the 73 cases with surgically resected bladder transitional cell carcinoma. The normal bladder tissue of twentytwo cases was used as control.The expression of Skp2 was tested by immunohistochemical method, and the related clinical phase and its expression were analyzed.Results: The expression rate of Skp2 was 50.7%. Skp2 expression was related with histological grade and pathological stage of tumors closely.Conclusion: Skp2 are important prognostic factors for patients with bladder transitional cell carcinoma.

膀胱移行细胞癌(BTCC)是泌尿系统最常见的恶性肿瘤,具有生长缓慢、多发性、复发性等临床特点。它的发生发展受多种基因的调控,在细胞周期中,G1—S期转换的调控是一关键点, Skp2和P27是近年来新发现细胞周期调控基因,主要参与G1—S期调控,是细胞从G1—S期转换的主要限速因子之一。Skp2是Fbox蛋白家族中重要的一员,通过泛素化降解P27蛋白,参与细胞周期调控,与肿瘤发生发展和预后密切相关[1]。本实验采用免疫组化技术对P27和Skp2在膀胱移行细胞癌及其癌旁或正常膀胱粘膜组织中的表达水平及相互关系,探讨Skp2与临床病理指标的相关性,评价对患者预后的价值。
1 材料与方法
1.1 一般资料
收集我院1998~2004年间手术切除后经病理确诊为膀胱移行细胞癌具有完整临床病理资料及组织标本共73例(TUR—Bt 51例、膀胱部分膀胱部分切除15例、根治性膀胱切除7例)病例。另外,选用22例非肿瘤粘膜供对照研究(其中12例为距肿瘤2cm 以上的癌旁正常粘膜,另10例为正常膀胱粘膜)。全部标本均进行P27和Skp2蛋
