CKIs蛋白在视黄酸诱导HL |

摘 要 从细胞周期调控这一途径探讨视黄酸抑制细胞增殖、诱导细胞分化的分子机制。结果显示:5×10-6mol/L全反式视黄酸(ATRA)及芳维甲(AE)处理的HL-60细胞生长受抑,4d后90%细胞停滞于G0/G1期,G1→S期转换受阻,同时NBT还原反应增强,Western Blot分析及免疫细胞化学染色表明处理后的细胞p16、p21两种细胞周期素依赖激酶抑制物(CKIs)表达增加,而CyclinD1和pRb蛋白则降低,其中尤以ATRA作用为明显。结果提示,细胞周期调控相关蛋白可能在视黄酸及其衍生物抑制细胞增殖、诱导细胞分化过程中起重要作用。 关键词 视黄酸 细胞周期素依赖激酶抑制物 细胞分化 中图号 R730.2
ROLE OF CKIs PROTEIN IN THE DIFFERENTIATION OF HL-60 CELL INDUCED BY RETINOIC ACID

Abstract In this report,the molecular mechanism of cellular proliferation inhibition and differentiation induced by retinoic acid was studied from the aspect of cell cycle control.The result showed,that 5×10-6mol/L all tran retinoic acid(ATRA)or arotinoidethylester(AE)could inhibit HL-60 cell growth and induce the cells differentiation,90 percent cells was arrested at G0/G1 phase after 4 days treatment of ATRA or AE,and the transition from G1 to S phase was blocked the expressions of p16 and p21 proteins,two kinds of cyclin dependent kinase inhibitors(CKIs),increased,but cyclinD1 and pRb proteins,the key regulators of G1→S phase transition,decreased.These results suggest that cell cycle relative proteins may play an important role in the control of cell proliferation inhibition and differentiation induced by retinoic acid and its derivatives. 视黄酸(Retinoic acid,RA)及其衍生物对多种肿瘤细胞具有增殖抑制和诱导分化作用,其中以对HL-60细胞的作用较为突出,但目前有关RA及其衍生物诱导细胞分化的分子机制仍不清楚。近年来,有关细胞周期调控分子机制的研究已有较大进展,比较一致的看法是细胞周期进程受一系列正和负信号的调节,细胞周期素(cyclins)、细胞周期素依赖性激酶(CDKs)、细胞周期素依赖性激酶抑制剂(CKIs)、p53、pRb蛋白以及某些癌基因的表达产物相互构成一个调控网络[1],此调控网络中的相关成分不但控制着细胞周期进程,同时也直接与细胞增殖与分化的调节相关联。上述调节信号的扰乱可能与以细胞恶性增殖为特征的生长失控、细胞低分化有重要联系[2]。本研究重点观察了全反式视黄酸(ATRA)及第三代视黄酸衍生物芳维甲(A[1] [2] 下一页 上一个医学论文: 血小板生成素转导鼠T淋巴细胞的增殖和分化 下一个医学论文: 井冈山市食品及公共场所从业人员乙肝表面抗原检测结果分析
