人胰腺导管上皮表达胰岛淀粉样多肽 |

【摘要】 目的 观察人胰腺导管上皮胰岛淀粉样多肽的分布。 方法 4例正常成人胰腺组织石蜡切片,用免疫组织化学PAP方法显示IAPP-IR细胞,Mayer苏木素复染。 结果 发现胰腺小叶间导管、小叶内导管、闰管上皮细胞至泡心细胞均呈胰岛淀粉样多肽免疫反应性,阳性物质主要分布于核上方及两侧。 结论 本研究证明人胰腺导管上皮细胞表达胰岛淀粉样多肽,推测其可能与胰腺的自我保护和胰液中碳酸氢盐的分泌有关。 【关键词】 胰岛淀粉样多肽;胰腺导管上皮;免疫组织化学;人 【中图分类号】 R329.1 【文献标识码】 A 【文章编号】 0529-1356(2000)03-283
EXPRESSION OF ISLET AMYLOID POLYPEPTIDE IN HUMAN PANCREAS DUCTAL EPITHELIUM

【Abstract】 Objective To observe the distribution of islet amyloid polypeptide(IAPP) in pancreatic ductal epithelium. Methods Four cases of normal human pancreases were obtained and immunohistochemical method and Mayer hematoxylin staining were used to show IAPP-immunoreative cells. Results It was found for the first time that IAPP distributes all the parts of ductal epithelium from interlobular duct to centroacinar cell and was located in the upside and both sides of nucleus. Conclusion The present work demonstrated that the human pancreas ductal epithelial cells express islet amyloid polypeptide, and supposed that IAPP may be related to the pancreas autoprotection and the secretion of HCO-3. 【Key words】 IAPP; Pancreas ductal epithelium; Immunohistochemistry; Human
胰岛淀粉样多肽(islet amyloid polypeptide, IAPP)是与胰岛素共存于正常胰岛B细胞同一分泌颗粒内的多肽类激素。近年来的研究发现,IAPP也存在于消化管等部位[1]。但关于IAPP在人胰腺导管上皮的分布国内外均未见报道。本研究就此进行了较为详细的观察,为进一步深入探讨IAPP的生理作用提供形态学依据。
材料和方法
收集4例正常人经Bouin液固定的胰腺石蜡块,切成5μm厚的连续切片,取相邻切片用Sternberger PAP法进行免疫组织化学染色,分别显示胰岛素(Ins)-和IAPP-免疫反应性(IR)细胞。主要步骤为:切片常规脱蜡到PBS,甲醇-H2O2和正常羊血清封闭后,分别滴加兔抗IAPP抗血清(1∶6 000, Peninsula Lab, USA产品)和豚鼠抗胰岛素抗血清(1∶600),4℃孵育过夜;羊抗兔IgG(1∶50)37℃ 40min;兔抗PAP复合物(1∶100)37℃ 40min;DAB显色盒(北京中山生物技术有限公司)显色,Mayer苏木精复染细胞核,常规封片。以PBS代替第一抗体作为阴性对照。
