与平滑肌细胞联合培养的内皮细胞的形态学和生长增殖 |

【摘要】 目的 探讨平滑肌细胞对内皮细胞形态和增殖的影响,为内皮细胞种植的组织工程学提供理论基础。 方法 模拟血管壁的内层结构及内皮细胞平滑肌细胞间的相互影响途径,建立内皮细胞与平滑肌细胞联合培养的模型,应用形态学、细胞计数对内皮细胞的结构和增殖进行研究。 结果 联合培养的新内皮细胞呈伸长性生长,沿细胞长轴排列的微丝增多,增殖受抑制。 结论 内皮细胞和平滑肌细胞联合培养的新模型是目前研究这两种细胞间相互影响的最佳方式。联合培养的内皮细胞的形态结构更接近于在体生理状态,这一变化可能更有利于其抗应力功能的调节。 【关键词】 血管;内皮细胞;平滑肌细胞;联合培养;形态学 【中图分类号】 R318.01;R318.11 【文献标识码】 A 【文章编号】 0529-1356(2000)03-274
MORPHOLOGY,STRUCTURE,GROWTH AND PROLIFERATION OF Ecs CO-CULTURED WITH SMCs

【Abstract】 Objective Effects of smooth muscle cells(SMCs)on morphology and function of endothelial cells (Ecs) has been studied in order to support the basic theory for tissue engineering. Methods A new co-culture model of Ecs-SMCs which mimicked the interior architecture of vessel and also imitated the interaction patterns between Ecs and SMCs was established. Morphological observation, cell calculating were applied in the research of morphology, proliferation of the Ecs. Results The results showed that the shape of co-culturing Ecs tended to elongating, the micro-filaments paralleling the cell membrane increased, the proliferation of the Ecs was inhibited. Conclusions The new co-culture model of Ecs-SMCs is the best one to reveal the interaction of the two kinds of the cells. The morphology of the co-culture Ecs is more similar to that in vivo. The change of the Ecs is beneficial to regulating the functions stress-resistance of the Ecs in co-culture model. 【Key words】 Vessel; Endothelial cells; Smooth muscle cells; Co-culture; Morphology
