血小板活化因子在颈髓损伤后线粒体功能损伤中的作用 |

摘 要 目的和方法:采用Allen打击法造成C6.7损伤,鞘内注射血小板活化因子(PAF)及静脉注射PAF受体拮抗剂NB52021,观察其对颈髓损伤后脊髓组织PAF含量、颈髓线粒体ATP酶活性、线粒体呼吸功能的影响。结果:颈髓损伤后颈髓组织PAF含量明显增加、蛛网膜下腔注射PAF可使伤后PAF含量增加更为显著;PAF能够抑制Ca2+-Mg2+-ATP酶、Na+-K+-ATP酶活性,明显降低线粒体耗氧量(OUR)、呼吸控制率(RCR)、磷氧比值(P/O)、氧化磷酸化效率(OPR):而BN52021能够抑制伤后颈髓组织PAF含量的增加,维持伤后线粒体Ca2+-Mg2+-ATP酶、Na+-K+-ATP酶活性的相对稳定,减轻线粒体功能损害。结论:PAF及其受体在颈髓损伤后颈髓线粒体功能损害的病理生理过程中起重要作用,PAF受体拮抗剂能够有效地保护颈髓损伤后线粒体功能。 主题词 创伤和损伤;线粒体;血小板活化因子;受体
Effects of platelet activating factor and its receptor on mitochondrial function damage after cervical cord injury

Abstract AIM and METHODS:C6,7 spinal cord injury models were performed in cats by Allen's method. Platelet activating factor (PAF) and PAF receptor antagonist BN52021 were administered by intrathecal or intravenous injection respectively. Their effects on PAF level, mitochondrial ATPase activity, mitochondrial function in cervical cord were observed after cervical cord injury. RESULTS:PAF levels in cervical cord tissue significantly increased following trauma. PAF levels were evidently elevated by arachnoid space injection of PAF. PAF receptor antagonist BN52021 could inhibit the increase of PAF levels in the cervical cord tissue following trauma; PAF could inhibit mitochondrial Na+-K+-ATPase, Ca2+-Mg2+-ATPase activities and evidently decrease mitochondrial oxygen uptake rate (OUR), respiratory control rate (RCR), P/O ratio and oxidative phosphorylation rate (OPR); PAF receptor antagonist BN52021 could sustain the relative stability of Na+-K+-ATPase, Ca2+-Mg2+-ATPase activity and improve mitochondrial respiratory function. CONCLUSION:PAF and its receptor play an important role in the pathophysiology process of mitochondrial function damage after cervical cord injury.
