摘 要 目的:探讨雷公藤红素对HMC-1细胞表达Fas、Fas-L的影响及与凋亡的关系。方法:用流式细胞仪检测细胞凋亡和Fas、Fas-L的表达。结果:HMC-1细胞能自发表达Fas,不表达Fas-L。经雷公藤红素作用,Fas呈先升后降的变化,Fas-L低水平表达。Fas阳性率下降与凋亡率增加相关。结论:Fas可能是雷公藤红素诱导HMC-1细胞凋亡的途径之一。

Abstract Objective:This paper reports the effect of tripterine on the expression Fas and Fas-L in HMC-1 cells and the relation to apoptosis.Methods:The expression of Fas/Fas-L and apoptosis were assayed by flow cytometry.Results:HMC-1 cells constituantly express Fas(22.7%),but not Fas-L.On exposure to tripterine at the concentration of 1.0 μmol/L,Fas positive rate increased to 57.5% at 2 h,then decreased to 12.5% at 4 h and 2.3% at 6 h.Fas-L expressed at lower level.There were significant relation between the decrease of Fas positive rate and increase apoptosis.Conclusion:Fas is one of the molecules which mediates HMC-1 cell apoptosis induced by tripterine.

雷公藤红素是雷公藤单体之一,属三萜类色素[1] 。它能诱导人肥大细胞系HMC-1细胞凋亡[2],但诱导凋亡的机制尚不清楚[3]。 Fas抗原(CD95)是一种膜受体,属TNFR/NGFR家族成员,表达于多种类型的细胞表面,其配体(Fas ligand ,Fas-L)主要表达于活化的淋巴细胞。Fas与Fas L结合能迅速地启动细胞凋亡。 本文研究雷公藤红素诱导HMC-1细胞凋亡过程中Fas 及Fas-L表达的变化及与凋亡的关系,以探讨雷公藤红素诱导HMC-1细胞凋亡的机制。
1 材料与方法
1.1 细胞培养 HMC-1细胞培养于5%CO2 , 37℃孵箱中。培养基IMDM(GIBCOBRL )含10% 热灭活小牛血清,0.1 U/L青霉素,0.1 U/L 链霉素。培养细胞密度为0.5×106~1.0×106 ml-1,取对数生长期细胞待用。
1.2 细胞凋亡的检测
1.2.1 形态观察 HMC-1细胞呈圆或椭圆形。凋亡时细胞膜内陷,核浓缩,形成凋亡小体。
1.2.2 流式细胞仪(FCM) DNA含量分析 1×106个细胞加50 μl含0.1% Triton-X100 的碘化丙啶(PI) (5 μg/ml) 染色,避光放置5 min,加0.5 ml PBS, 上流式细胞仪检测,Multicycles软件分析结果。
