免疫学技术与方法流式细胞术定量检测细胞凋亡3种方法的比较研究 |

摘 要 目的：探讨流式细胞术定量检测细胞凋亡3种方法的价值。方法：同时使用PI染色、TUNEL及Annexin V/PI 3种方法定量检测地塞米松处理小鼠的胸腺细胞凋亡发生率。结果：PI染色、Annexin V/PI、TUNEL 3种方法凋亡检出率分别为27.19%、32.28%、50.17%，两两之间均有显著差异(P＜0.01)。但仅Annexin V/PI法可将正常细胞、凋亡细胞、死亡细胞区分开来。结论：Annexin V/PT法是目前最为理想的凋亡定量检测方法。

Abstract Objective:Evaluation of three methods for measuring apoptosis by flow cytometry.Methods:The apoptotic percentage of dexamethason-treated mouse thymocytes was measured by flow cytometry using propidium iodide(PI) staining.TUNEL and Annexin V/PI double staining respectively. Results：The apoptotic percentage measured by PI staining,Annexin V/PI and TUNEL assays were 27.19%,32.28% and 50.17% positive respectively.There are significant difference among the results.But only the Annexin V/PI assay can discriminate normal cells,apoptotic cells and necrotic cells quite well.Conclusion：The Annexin V/PI assay is the best method for measuring apoptosis by flow cytometry presently.

细胞凋亡的定性检测依赖于形态学观察及DNA电泳,其定量检测则需借助于流式细胞检查。使用碘化丙啶(PI)染色检测DNA含量是最早出现的凋亡定量检测方法［1］。进入90年代,检测DNA断裂点的TUNEL(Terminal deoxylnucleotidyl transferase mediated-dUTP nick end labeling)技术成为凋亡定量检测的主流。1995年Vermes 首次使用荧光素标记的膜联蛋白V(FITC- Annexin V)联合PI染色双参数技术定量检测凋亡，目前国内尚未见报道［2］。我们同时使用上述3种方法对地塞米松处理的小鼠胸腺细胞进行凋亡检测对比研究，借以探讨这一新的方法的应用价值，现报道如下。
1 材料与方法
1.1 材料 4周龄雄性昆明小鼠12只。PI(Sigma)。TUNEL试剂盒(德国Boehringer Mannheim)。Annexin V/PI试剂盒(法国国际免疫公司)。流式细胞仪(FACScan型 美国BD公司)。
1.2 方法
1.2.1 动物模型及细胞制备 10只小鼠腹腔注射地塞米松25 mg/kg，2只小鼠腹腔注射生理盐水作为对照。10 h后取胸腺，网搓法分离胸腺细胞。PBS洗2遍后调细胞浓度2×106 ml-1，备用。
1.2.2 PI染色法 取2×106 细胞,加入80%冷乙醇，4℃ 30 min，PBS洗1次，加入P[1] [2] [3] 下一页
