免疫调节剂对大鼠巨噬细胞脂质过氧化物和酸性磷酸酶的影响 |

摘要 目的和方法:用生理盐水灌洗大鼠的腹腔和肺泡,收集腹腔与肺泡巨噬细胞,37℃培养2 h后测定脂质过氧化物的含量和酸性磷酸酶的活力。结果:腹腔和肺泡巨噬细胞内脂质过氧化物的水平与酸性磷酸酶的活力均可被环磷酰胺和细脚拟青霉所调节,其中环磷酰胺使脂质过氧化物水平升高,酸性磷酸酶活力降低,细脚拟青霉则反之,使脂质过氧化物水平下降,酸性磷酸酶活力升高。结论:巨噬细胞内脂质过氧化物水平与酸性磷酸酶活力呈负相关,且可被免疫抑制剂和免疫激活剂所调节。 主题词 过氧化脂质类; 酸性磷酸酶; 巨噬细胞; 佐剂,免疫
Effects of immunomodulators on lipid peroxides level and acid phosphatase activity in macrophages of rats

Abstract AIM and METHODS:The relation and regulation of lipid peroxides (LPO) level and acid phosphatase (ACP) activity in peritoneal macrophage (PMΦ) and alveolar macrophage (AMΦ) of rats were studied. After PMΦ and AMΦ of rats were irrigated by normal saline and collected by centrifuge and 2 h in a humidified incubator at 37 ℃, LPO level and ACP activity in PMΦ and AMΦ were detected by biochemical methods. RESULTS: The ACP activity was decreased, while the LPO level was increased in the PMΦ and AMΦ of rats administered with cyclophosphamide. However, the ACP actvity was increased, while the LPO level was decreased in the PMΦ and AMΦ of rats administered orally with paecilomyces tenuipes water extract .CONCLUSION: The level of LPO was negatively related to the activity of ACP, and it was regulated by immunomodulators. 脂质过氧化物(lipid peroxides, LPO)被认为是由细胞膜相结构中的多不饱和脂肪酸过氧化而产生的,有研究报道LPO可能是一种非特异性的免疫抑制剂[1]。酸性磷酸酶(acid phosphatase, ACP)被认为是巨噬细胞(macrophages, MΦ)的标志酶,ACP活力的高低反映了MΦ被激活的程度[2]。本文拟研究大鼠腹腔巨噬细胞(perritoneal macrophage, PMΦ)和肺泡巨噬细胞(alveolar macrophage, AMΦ)内LPO水平与ACP活力的相关性,以及免疫抑制剂环磷酰胺与免疫激活剂细脚拟青霉(paecilomyces tenuipes,PT)对LPO水平与ACP活力的调控作用。
