低剂量辐射增强小鼠胸腺细胞PKC亚型的表达 |

摘 要 目的和方法:本实验采用流式细胞术研究了不同剂量X射线全身照射后4、12、24和48h,昆明小鼠胸腺细胞蛋白激酶C亚型表达的变化。结果:75mGy全身照射后PKC-α、β1、β2表达均有增强,于照射后12h达峰值,PKC-α、β2于48h恢复至假照水平,PKC-β1于48 h仍明显增高。4Gy照射后,PKC-α表达显著下降。 主题词 辐射剂量;蛋白激酶C;流式细胞术;胸腺
Increased expression of PKC isoforms in thymocytes after WBI with low dose X-rays

Abstract AIM and METHODS: Male Kunming mice were whole-body irradiated with 75 mGy or 4 Gy X-rays. The expressions of PKC (protein kinase C) isoforms of thymic lymphocytes were analyzed by immuno-fluorescence flow cytometry with corresponding polyclonal antibodies at different time intervals after irradiation. RESULTS: The positive cells for PKC α,PKC β1 and β2 all increased after WBI with 75 mGy, reaching its peak 12 hours after WBI. The expressions of PKC α,β2returned to normal level, and the expression of PKC β2 still remained at a higher level than normal 48 hours after WBI with 75 mGy. The positive cells for PKC α decreased 24 and 48 hours after WBI with 4 Gy. CONCLUSION: Low dose radiation can induce expression of PKC isoforns and promote signal transduction of T lymphocytes. 近几年来,大量的实验使人们逐步认识到低剂量辐射能够引发一些高剂量辐射所致不同的生物效应,已成为辐射研究领域的一个新的热点。其作用机制正不断被揭示[1]。就淋巴细胞信号传递系统而言,本实验室报道75mGy X射线单次全身照射后,小鼠TCR/CD3的表达增强,c-fos,c-jun表达上调,细胞内[Ca2+]i动员加快,PKC总活性增高[2]。对蛋白激酶C(protein kinase C,PKC)亚型的研究尚未见报道。本实验应用流式细胞术,观察低剂量照射后不同时间PKC-α,PKC-β1及PKC-β2蛋白表达的变化,进一步分析低剂量辐射对信号传递的影响。
材料与方法 一、实验动物: 雄性昆明系小鼠20±2g,购于白求恩医科大学实验动物部。设75mGy、4Gy照射组和假照组,照射后4、12、24、48h断头处死小鼠取胸腺,制单细胞悬液。每组15只鼠,每3只小鼠的胸腺细胞混合为一样本,即每组n=5。 二、照射条件: Philips深部X射线全身照射(WBI),电压200kV,电流10mA,滤过板0.5mm铜,1.0mm铝,球靶距50cm或212cm,总剂量为4Gy(剂量率0.287Gy/m[1] [2] [3] 下一页 上一个医学论文: 甲硫 脑啡肽对脾交感神经活动的双向调节作用 下一个医学论文: 内皮素在急性肝衰竭时肾功能障碍发生中的作用
