烫伤大鼠室旁核和触液神经元中胆囊收缩素的变化

  【摘要】 目的 明确胆囊收缩素在烫伤应激中的确切作用。 方法 在大鼠清醒状态下给予标准的25%Ⅲ度皮肤烫伤,伤后24h杀动物,用免疫细胞化学ABC法显示PVN和CSF-CNs的CCK阳性神经元,结果用显微图像分析技术处理。 结果 (1)烫伤大鼠PVN代表平面CCK神经元的总面积增大,免疫染色无明显变化。(2)正中隆起(medin eminence,ME)的CCK免疫染色阳性纤维的面积密度无显著性差异而免疫染色减弱。(3)烫伤大鼠第三脑室室管膜内CCK免疫染色阳性的CSF-CNs数量、面积和免疫染色强度均高于对照组。 结论 烫伤应激使CCK在胞体的合成和末梢的释放均增加。烫伤后大量CSF-CNs的出现为CCK释放入脑脊液提供了一条新的途径。CCK可能通过脑脊液循环来调节控制垂体的肽能神经元活性以参与对烫伤应激的调节。

NUCLEUS AND CEREBROSPINAL FLUID-CONTACTING
NEURONS AFTER SCALDING IN RAT

  【Abstract】 Objective The cholecystokinin (CCK) plays an important role in regulating various stress.The paraventriclar nucleus (PVN) and cerebrospinal fluid-contacting neurons(CSF-CNs),which may participate in the responding to scaled,contain some CCK.There have been hints that CCK is involved in scalding stress.The aim of the present study is to examine the role of CCK which is in the PVN and CSF-CNs during scalding stress.Methods Adult male Wistar rats were given a standard scald on 25% of the body surface or only a sham.They were sacrificed at the 24th hours after scald.The results were showed with ABC immunocytochemical method and examined with microscopic image analyse system at the light microscope level.Results It showed that there were some changes of CCK in the PVN and CSF-CNs.However there was no difference of the immunostaining intensity in PVN and no difference of area density in medin eminence (ME) between the burnt and the sham rats,the areas of CCK-positive neurons in PVN are larger and the CCK immunostaining intensity of the neurites in ME was lighter in burnt rats than that in control
