脊髓混合性胶质细胞机械损伤后的形态变化

  【摘要】 目的 建立体外培养的脊髓胶质细胞机械损伤模型,观察机械性损伤刺激后,混合性胶质细胞内不同细胞的形态变化及其特性。 方法 14~15d胎龄的小鼠脊髓制成细胞悬液后,在含10%胎牛血清的DMEM培养液中培养4周,形成富含星形胶质细胞的平铺混合胶质细胞,用塑料滴头刮损后,在刮损缘的反应性胶质细胞增生,肥大,向刮损裸区内生长并发生细胞分裂。 结果 增长的细胞沿刮损方向呈极性排列,可伸出纤长的突起,突起上有串珠样结构自胞体向突起末端移动,并可相互间发生融合。 结论 免疫细胞化学染色表明损伤边缘的胶质细胞GFAP染色加深,但进入损伤区的细胞并非GFAP阳性,也不为识别纤维细胞抗原的抗体所标记,其可能的来源和特性还有待进一步确定。

MORPHOLOGICAL CHANGES OF CULTURED SPINAL CORD
MIXED GLIAL CELLS AFTER MECHANICAL INJURY

  【Abstract】 Objective To investigate the response of embryo spinal cord glia cells to mechanical injury in mixed glia culture of E14-15d mouse embryo spinal cord.Methods The E14-15d mouse embryo spinal cord was dissected in sterile condition,and cultured in incubator at 37℃5%CO2 for 4 weeks to obtain the mixed glia cells enriching astrocytes.A mechanical scratch was done on the cell layers by a plastic tip.Phase contrast microscopic photos were taken every 10 min for 48hrs.The cells were then stained by ABC immunocytochemistry method for antibodies against astrocytes marker GFAP,oligodendrocytes marker GC,neuron specific NSE and fibroblast specific Thy1.1.Results About 4 hours after the injury,cells started to invade from the scratch edge into the blank region.These cells had long processes.Vesicles were seen moving from the cell body distally.The vesicles could fuse with each other.The processes were found to elongate and retract rapidly.Mitosis could be seen sometimes.NSE and Thy1.1,or GFAP could label none of the cells invaded to the blank region.Some could be recognized by GC,whereas the GFAP was stained heavily at the edge.Conclusion The cells were found migrating actively into the scratched region.The characteristics of these cells remains to be identified.Presumably,astrocytes would be the most active elements.However,no
