ETB受体激活对猪冠状动脉阻力血管内径的影响 |

摘 要 目的:观察ETB受体激活对猪冠状动脉阻力血管有何影响。方法:采用电视显微镜观察离体猪冠状动脉阻力血管内径。实验分为3组。各组分别在对照观察、内皮破坏或10-5mol/L L-N-硝基精氨酸甲酯(L-N-nitro-argining methyL ester,L-NAME)处理后,给予内皮素B受体(endothelin-B receptor,ETB)激活剂sarafotoxin 6c(SF6c),记录浓度-反应曲线。结果:给予SF6c后,对照组血管呈现浓度依赖性轻度舒张及轻度收缩反应:内皮破坏组及一氧化氮合成酶抑制组仅表现为浓度依赖性收缩反应。结论:ETB受体激活可对冠状动脉阻力血管产生收缩舒张双重作用;其中舒张作用依赖内皮存在和一氧化氮的产生。 主题词 内皮缩血管肽类;冠状血管;血管阻力
Endothelin-B receptor stimulation in porcine coronary resistance arteries

Abstract AIM:To Study the relative role of ETB receptor stimulation in porcine coronary resistance arteries.METHODS:Isolated porcine coronary resistance arteries were studied using in vitro videomicroscopy. Dose-response curves to the ETB agonist sarafotoxin 6c (SF6c) were determined under control conditions,after endothelial denudation or after 10-5 mol/L L-NAME.RESULTS:In control vessels,SF6c caused a weak vasodilation that followed a constriction. After endothelial denudation or blocking NO synthase with L-NAME, however, the vasodilation effects of SF6c did not appear except the dose-dependent constriction.CONCLUSION:The weak vasodilation and vasoconstriction can be induced by endothelin-B receptor stimulation in coronary resistance vessels, and the effect of relaxation of coronary resistance arteries due to ETB receptor stimulation is dependent on endothelium and mediated by NO. 内皮素(endothelin,ET)是一种重要的血管活性多肽,它通过内皮素受体发挥作用。目前已知内皮素至少有两种受体,ETA和ETB受体。过去一直认为ETB受体激活主要引起内皮依赖性血管舒张。然而近期有人报道ETB受体也参于某某些血管的收缩。目前已知,冠状动脉上也存在ETB受体[1],Bax等[2]和Teerlink等[3]分别报道高浓度ETB受体激动剂可引起冠状动脉大血管的收缩。但对冠状动脉阻力血管作用如何,尚不清楚。本工作给予猪冠状动脉阻力血管(<200μm)以ETB受体 激动剂sarafotoxin 6c (SF6c),观察ETB受体在冠状动脉阻力血管中的作用。
材料与方法
