T细胞无能与凋亡关系初探 |

摘 要 目的:探讨无能T细胞与凋亡的关系。方法:利用B7-1单抗和环孢素A(CsA)联用处理APC即T细胞系统,在体外诱致抗原特异性T细胞无能,用PI染色法经FACS分析了无能T细胞的凋亡,用RT-PCR法检测了凋亡细胞Fas mRNA的表达水平。结果:无能T细胞在1、3、5、10 d细胞凋亡的百分率依次为2.16%、11.28%、19.27%、41.22%。当加入100 U/ml IL-2维持培养,无能T细胞未显示明显凋亡。无能T细胞活化后经PHA、PMA+A23187、CD3单抗再次诱导,24 h便发生明显凋亡,凋亡百分率分别为34.64%、21.27%、15.07%,活化后的T细胞及无能T细胞凋亡时Fas mRNA上调表达。结论:无能T细胞在体外是逐渐发生凋亡的,Fas/FasL参与细胞凋亡过程。

Preliminary study on the correlation between T cell anergy and apoptosis

Abstract Objective:Study the correlation between T cell anergy and apoptosis.Methods:Antigen-specific T cell anergy was induced by the combination B7-1mAb and cyclosporinA(CsA) in vitro.Anergic T cell apoptosis was analyzed by FACScan with PI-staining. Fas mRNA expression in apoptic cells was detected with RT-PCR. Results:Apoptic percentage of anergic T cells were 2.16%,11.28%,19.27%,41.22%during 1 d,3 d,5 d,10 d.Evident apoptosis was not observed in the anergic T cells cocultured with 100 U/ml IL-2. PHA-activated anergic T cells reactivated by PHA,PMA+A23187 and CD3mAb showed significant apoptosis at 24 h, their apoptic percentages were 34.64%,21.27%,15.07%,respectively.Fas mRNA dramatically up-regulated expression in T cells and anergic T cells activated with the above stimuli. Conclusion:Anergic T cell was gradually apoptic in vitro Fas/Fasl interaction participated in the apoptosis.

自身反应性T细胞在胸腺内排除是不完全的,在成熟淋巴细胞中存在着自身反应性T细胞,机体仍处于免疫自稳状态,表明成熟T细胞是可以产生免疫耐受的。这些无能T细胞是一直存在,还是被排除,目前尚未见研究报道。Daniel等发现转染B7的肿瘤细胞可阻止CTL发生凋亡,推断缺乏共刺激信号,能够增强T细胞凋亡的敏感性[1]。研究报道,凋亡不仅局限于未成熟或转化的T细胞,在表达TCRαβ或γδ的成熟T细胞亦可发生凋亡[2,3]。又有报道淋巴细胞激活诱导后,除伴有细胞因子产生和增殖外,同时可发生激活诱导的细胞死亡(Activation-induced cell death,AICD)[4,5]。AICD可能是外周反应性T细胞被排除的机制,在[1] [2] 下一页 上一个医学论文: 创伤后巨噬细胞抑制T细胞功能的分子机制研究 下一个医学论文: 香菇多糖注射液对反复呼吸道感染患者免疫功能的影响
