肺动脉与冠状动脉内皮环氧合酶及TXA2合酶基因表达的差异

  摘 要 目的与方法:应用分子杂交和放免法观察了缺氧下培养的猪肺动脉内皮细胞(PAEC)和冠状动脉内皮细胞(CAEC)的环氧合酶-1(COX-1),环氧合酶-2(COX-2)和血栓素A2合酶(TXS)基因表达及前列腺素生成的变化。结果:(1)常氧下培养24 h PAEC的COX-2 mRNA含量为CAEC的1.5倍;PAEC的PGI2生成量明显高于CAEC。(2)缺氧培养24 h PAEC和CAEC中COX-1mRNA均增加,分别是其常氧对照组的2.5倍和3倍,而COX-2 mRNA含量仅在PAEC中明显增多,达其对照组的1.6倍。(3)缺氧对CAEC的PGI2生成量增加的幅度大于PAEC的增量。(4)PAEC和CAEC在常氧下和缺氧时TXS基因表达TXA2产量均无明显差异。结论:PAEC和CAEC在常氧下和缺氧时COX基因表达和PGI2生成的差异可能与肺动脉和冠脉的基础张力和缺氧反应的差异有关。

Expression of cyclooxygenase and thromboxane synthase genes in pulmonary and coronary arterial endothelial cells

  Abstract AIM:The expression of cyclooxygenase-1 (COX-1), cyclooxygenase-2(COX-2) as well as thromboxane synthase (TXS) genes and the production of PGI2 and TXA2 in cultured pulmonary and coronary artery endothelial cells (PAEC and CAEC) were studied by methods of dot blot and RIA.RESULTS:(1)After cultured in normoxia for 24h, COX-2 mRNA in PAEC was 1.5 times as much as those in CAEC(P<0.05); (2)After cultured in hypoxia for 24 h, COX-1 mRNA were 2.5 times in PAEC and 3 times in CAEC compared with normoxic control. However, COX-2 mRNA only increased in hypoxic PAEC, which was 1.6 times that of it s normoxic group; (3) Hypoxia increased the production of PGI2 both in PAEC and CAEC, its increment in PAEC was higher than that in CAEC;(4) There was no difference in TXS gene expression and TXA2 production of PAEC and CAEC either in normoxic or in hypoxic group.CONCLUSION:The difference in COX gene expression and PGI2 production between PAEC and CAEC might be responsible for their different basal tone and their different responsiveness to hypoxia.
