实验性隐睾诱导小鼠生精细胞凋亡的研究 |

【摘要】 目的 研究手术隐睾所致成年小鼠生殖细胞凋亡及相关调节因子bcl-2、bax蛋白在曲细精管中的定位、变化。 方法 以末端脱氧核糖核酸转移酶介导的dUTP缺口末端标记法(terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling method,TUNEL)检测凋亡的生殖细胞,生物素-抗生物素蛋白DCS(biotin-avidin D cell sorter)体系间接免疫荧光法检测Bcl-2、Bax蛋白在曲细精管中的定位、分布。 结果 隐睾术后3d,手术侧睾丸重量及细胞凋亡数量与对侧无明显区别。而6~15d,睾丸重量明显减轻,凋亡细胞明显增多,以精母细胞、精子细胞为主。在正常小鼠曲细精管中,bcl-2大量表达,呈散在的颗粒状存在于各级生精细胞中,以精子细胞胞质为主。Bax蛋白表达量较少,一部分也以散在的颗粒存在于各级生精细胞质中,另一部分则较致密地分布于曲细精管基底部。隐睾术后3d,Bcl-2、Bax的表达与对侧无明显区别;6~15d,Bcl-2明显下降,Bax明显增加。 结论 隐睾明显诱导生殖细胞凋亡,Bcl-2、Bax蛋白对生殖细胞的凋亡有重要调节作用,提示温度、Bcl-2、Bax蛋白在精子发生过程中对生精细胞的主动退化可能有重要作用。 【关键词】 生精细胞凋亡 Bcl-2蛋白 Bax蛋白
CRYPTORCHIDISM INDUCES TESTICULAR GERM CELL APOPTOSIS IN MICE TESTES AND CHANGES OF Bcl-2, Bax PROTEIN EXPRESSION

【Abstract】 Objective To investigate the effect of experimentally induced cryptorchidism on apoptosis in testes of adult mice and the distribution changes of Bcl-2, Bax protein. Methods Total DNA was isolated from testes and was electrophoretically separated in 1.5% agarose gel, TUNEL method, biotin-avidin DCS system indirect immunofluorescence techniques were used. Results DNA fragmentation, increasing apoptotic germ cells, in coordination with the loss of testicular weight, were observed on 6-15d in the cryptorchid testes. By TUNEL staining it was noted that postive cells were most frequently among primary spermatocytes and round spermatids. Bcl-2, Bax protein mainly distributed in cytoplasm of germ cells. With the increase of apoptosis, Bcl-2 was down and Bax was up regulated. Conclusion Temperature, Bcl-2, and Bax protein play an important role in male germ cell apoptosis. 【Key words】 Apoptotic germ cells; Bcl-2 protein; Bax protein
