枸杞多糖体外对正常及衰老小鼠免疫细胞功能的作用及作用机理的初步研究 |

摘 要 目的:探讨枸杞多糖(LBP)对免疫细胞功能的直接作用及其机理。方法:应用淋巴细胞增殖、溶血空斑和Fura-2/AM双波长荧光测定等方法。结果:LBP体外应用对正常小鼠及快速老化模型SAMP8和抗快速老化模型SAMR1脾细胞增殖反应具有直接促增殖作用,明显增加正常小鼠脾抗体形成细胞(PFC)数目,迅速诱导脾细胞和腹腔巨噬细胞内[Ca2+]i的增加,并有一定剂量依赖性关系。结论:枸杞多糖对免疫活性细胞的功能有直接促进作用,这可能与其增加细胞[Ca2+]i有关。 关键词 枸杞多糖 淋巴细胞增殖反应 抗体形成细胞 游离Ca2+ Fura-2/AM 中国图书分类号 R967
Effects of lycium barbarum polysaccharides on the splenocyte immune function of normal and aged model mouse and it s possible mechanism in vitro

Abstract Objective:To explore the action of immuno-modulation of lycium barbarum polysaccharides(LBP) on mouse splenocyte and it s possible mechanism in vitro.Methods:3H-TdR incorporation plaque-forming cell(PFC) assays and Fura-2/AM fluorescence double-wave measurement.Results: LBP not only enhanced splenocyte proliferation of LACA,senescence-accelerated mouse(SAMP8)and antisenescence-accelerated mouse(SAMR1)directly,but also increased the number of PFC of LACA mouse splenocytes.Furthermore,the concentration of free Ca2+ in splenocytes and macrophages was increased rapidly with dose-depend manner induced by LBP.Conclusion :LPB possesses direct immunopromoting effect on immune cell function and this effect maybe related with increasing concentration of free Ca2+ in cytoplasm,[Ca2+]i. 枸杞是传统的名贵补益中药,已有几千年的药用历史,具有“滋阴补血、益精明目”之功效。近年来,我室与植化室密切合作从中分离获得了枸杞多糖(lycium barbarum polysaccharides,LBP,编号901119)。免疫药理学研究表明,枸杞多糖体内应用不仅增强正常小鼠机体的免疫功能,而且对去胸腺、D-半乳糖衰老模型小鼠及自然衰老小鼠的免疫功能低下具有明显的改善作用[1-3],但对其作用的分子机理研究较少。本文研究发现,LBP在体外直接促进正常小鼠和衰老模型小鼠脾细胞增殖反应,具有原丝分裂原样作用。随后从细胞信号传导途径开始研
