葡萄糖调节蛋白94在人胚胎发育中的表达 |

【摘要】 目的 为探讨葡萄糖调节蛋白94(GRP94)在胚胎发育中的作用,研究了10~31周人胎组织GRP94的定位及表达。 方法 用SP免疫组织化学方法。 结果 结肠粘膜上皮、肾小管上皮及肝细胞的胞质内,在10周已有GRP94的表达,18~20周时表达增多,一直维持较高水平。胃粘膜上皮及心肌细胞也有较强的表达。肺、骨骼肌、脊髓及胸腺未见表达。 结论 GRP94存在着基础表达,可能在某些器官的发育过程中起作用。 【关键词】 葡萄糖调节蛋白94 热休克蛋白 胚胎发育
STUDY ON THE EXPRESSION OF GLUCOSE-REGULATED PROTEIN94 IN DEVELOPMENT OF THE HUMAN FETUSES

【Abstract】 Objective Glucose-regulated protein94(GRP94),which belongs to HSP90 family, is resided to the endoplasmic reticulum(ER) as molecular chaperones. We examined the distribution of GRP94 development human embryonic organs.Method 9 human fetuses(weeks 10-31 of gestation)were studied by immunohistochemistry.Results The expression of GRP94 had been shown in the epithelial cells of colonic mucosa,renal tubule and hepatocytes,localized in the cytoplasma of the cells.GRP94 was observed at gestational week 10,increased markedly from 18-20 week and maintained at constantly high level.It was also detected in the gastric mucosa and cardiac muscle.No GRP94 expression was shown in lung、skeletal muscle、spinal cord and thymus.Conclusions GRP94 is constitutively expressed within specific tissues and suggest that GRP94 might play an important role in the embryogenesis. 【Key words】 Glucose-regulated protein 94; Heat shock protein; Embryogenesis
葡萄糖调节蛋白(glucose-regulated proteins,GRP94)是一类应激蛋白,与热休克蛋白(heat shock proteins,HSPs)有较高的同源性,其中常见的GRP94和GRP78位于内质网腔,作为分子伴侣协助蛋白质的正确折叠和装配。特别是GRP94由内质网合成,为驻留于内质网腔的内质网标志分子伴侣,具有多种重要的生物学功能[1,2]。最新研究发现,GRP94在小鼠心脏的器官发生中有作用[3]。迄今,有关HSPs对胚胎发育的作用已有大量研究,但对GRPs的了解却很少,特别是在人胎中的有关研究国内外未见报道。鉴于此,我们应用免疫组织化学方法,探讨了GRP94在胚胎组织中的表达及定位。
材料和方法
