实验性脾虚证大鼠胰岛细胞的免疫组织化学研究 |

【摘要】 目的 研究实验性脾虚证大鼠胰岛细胞胰岛素、胰岛淀粉样多肽、胰高血糖素和生长抑素分泌活动的变化,探讨中医学脾虚证发病与胰岛激素分泌活动的关系。 方法 用Wistar雄性成年大鼠24只,分为正常对照组、实验性脾虚组、自然恢复组和中药治疗组。4组动物同时麻醉处死,取胰尾固定于Bouin液,制成石蜡切片。用免疫组织化学法显示胰岛B细胞胰岛素和胰岛淀粉样多肽、A细胞胰高血糖素和D细胞生长抑素,并用显微分光光度计对免疫染色强度进行了定量测定。 结果 与对照组相比,脾虚组胰岛B细胞胰岛素和A细胞胰高血糖素免疫染色明显减弱,而D细胞生长抑素和B细胞胰岛淀粉样多肽免疫染色明显增强。 结论 上述结果提示,胰岛细胞激素分泌活动失常可能与脾虚证代谢异常有密切关系。 【关键词】 胰岛细胞 实验性脾虚证 免疫组织化学 大鼠
IMMUNOHISTOCHEMICAL STUDY ON THE PANCREATIC ISLET CELLS IN EXPERIMENTAL SPLEEN DEFICIENCY SYNDROME OF RATS

【Abstract】 Objective To study the relationship between the islet hormonal secretory activity and the pathogenesis of spleen deficiency syndrome in Chinese traditional medicine.Methods The changes of secretory activity of insulin(Ins), islet amyloid polypeptide (IAPP),glucagon(Glu) and somatostatin (SOM) in pancreatic islet cells of experimental spleen dificiency syndrome rats were observed by immunohistochemical technique.24 adult male Wistar rats were divided into 4 groups:(1)normal control group; (2)experimental spleen deficiency syndrome group induced by rhubarb combined with imbalance of food supply;(3) therapeutic group treated with Chinese herbs-“Plus Si Jun Zi Decoction”,and (4) self-recovery group,that is,the syndrom was induced but without medical treatment.The tail portions of pancreas were taken and prepared for immunohistochemical demonstration of A cells (for Glu),B cells(for Ins and IAPP) and D cells (for SOM).The immunoreactivity of the above mentioned cells were measured by microspectrophotometry.Results As compared with the normal control,the immunereactivity of Ins in B cells and Glu in A cells of the experimental spleen deficiency syndrome group is obviously declined, while IAPP in B cel[1] [2] 下一页 上一个医学论文: 鸡胚房间隔与第二孔的形态发生 下一个医学论文: 金属硫蛋白与心肌血管内皮细胞的特异结合
