人肿瘤浸润性树突状细胞的表型分析及原因探讨 |

中国图书分类号 R73-3 摘 要 目的:树突状细胞作为抗原提呈细胞在机体的抗肿瘤免疫反应中起重要作用,对肿瘤浸润性树突状细胞的研究有助于揭示肿瘤免疫逃逸机制。方法:采用酶消化、细胞分离、流式细胞仪分析等方法研究了14例胃肠道肿瘤患者肿瘤浸润性树突状细胞的表型,并用RT-PCR检测了IL-10、VEGF和TGF-β13种免疫抑制因子在14例肿瘤细胞中的表达。结果: 肿瘤浸润性单核细胞中有CD1a+的树突状细胞存在,该类细胞的表面分子如MHC-II类分子、CD80及CD54等共刺激分子和粘附分子有低表达或不表达,14例肿瘤细胞中IL-10、VEGF及TGF-β1的表达率均较高。结论:肿瘤浸润性树突状细胞可能是一种无功能或功能低下的树突状细胞,肿瘤细胞在肿瘤局部分泌的免疫抑制因子可能是导致肿瘤浸润性树突状细胞功能低下的原因之一。 关键词 树突状细胞 肿瘤浸润 MHC-II类分子 IL-10 VEGF TGF-β1 免疫抑制
The phenotypes of human tumor infiltrating dendritic cells in gastrointestinal cancers

Abstract Objective: As antigen-presenting cells,dendritic cells(DCs) p?ay important roles in antitumor immune response.Analysis of tumor infiltrating dendritic cells(TIDCs)would contribute to investigate the mechanisms by which tumors escape immune recognition.Methods:Phenotypes of TIDCs were examined by methods including digestion with enzyme,cell separation and FACS.Reverse transcript-polymerase chain reaction(RT-PCR)was used to detect IL-10、VEGF and TGF-β1 mRNA expression in the freshly isolated tumor cells of 14 patients.Results:The expression of some molecules such as MHC class II and costimulating factors(CD80),adhesion molecules(CD54)on the surface of TIDCs were downregulated or negative .Levels of IL-10、VEGF and TGF-β1 mRNA expression in tumor cells of 14 patients were very high.Conclusion:The results suggested that TIDC was a kind of dysfunctional DC and immunosuppressive factors from tumor cells could inhibit DC s function .
