SLE患者TCR V 基因亚家族的表达特征及其克隆性 |

中国图书分类号 R593.24 R392.11 摘 要 目的:探讨SLE患者外周血淋巴细胞中TCR Vβ基因亚家族的表达及其克隆性。方法:采用RT-PCR-Southern杂交分析TCR Vβ 1~20基因亚家族的表达水平,进而用RT-PCR扩增TCR Vβ 24个亚家族的CDR3,并经基因扫描分析确定T细胞的克隆性。结果:6例SLE患者Vβ 8和Vβ 16的表达明显增高, 2例患者的Vβ 2和Vβ 9产物来源于寡克隆T细胞。结论:在自身抗原刺激下,TCR Vβ基因存在着优势取用; T细胞的克隆性增殖则可能是由特定的自身抗原所引起的应答。 关键词 系统性红斑狼疮 TCR Vβ基因 T细胞克隆性
Characteristics of TCR Vβ gene subfamily expression and T cell clonality in patients with systemic lupus erythematosus

Abstract Objective:To investigate the TCR Vβ gene subfamily expression and T cell clonality in patients with Systemic lupus erythematosus (SLE). Methods:The level of TCR Vβ gene expression was analyzed using RT-PCR and Southern blotting. CDR3 regions of TCR Vβ 1~24 gene were amplified by RT-PCR, then analyzed by Genescan in order to determine the T cell clonality. Results:Significant increases were found in the level of expression of Vβ 8 and Vβ 16 gene , Morever ,Vβ 2 and Vβ 9 products were derived from oligoclonal T cells. Conclusion:The results suggested that the usage of TCR Vβ gene was restricted when stimulated by some autoantigens and that the clonal expansions of T cells may be a specific autoantigen-driven response. 系统性红斑狼疮(SLE)患者多伴有细胞及体液免疫异常, 最终导致自身抗体的产生。在细胞免疫异常中,T细胞的作用显得更为重要,因为T细胞能调节B细胞的功能,而且在SLE中大多数致病性自身抗体是T细胞依赖性的[1]。T细胞受体(TCR)是T细胞识别抗原及被激活的中心环节,TCR的选择性表达可能与某些炎症性风湿病的免疫发病机理有关[2]。TCR β基因有47个有功能的V基因片段,根据核苷酸顺序同源性大于等于75%的原则,其分属26个家族[3]。本研究对SLE患者的TCR Vβ基因亚家族的表达特征及其克隆性进行了分析。
1 材料与方法
