U46619对培养的人气道平滑肌细胞增殖的作用 |

摘 要 目的和方法:研究U46619(9,11-dideoxy-11α,9α-epoxymethano-prostaglandin F2α)对培养的人气管平滑肌细胞增殖的作用。分离人的气管平滑肌细胞并且进行传代培养。在培养基中加入各种浓度的U46619,计数细胞并且测定[3H]-胸腺嘧啶核苷([3H]-TdR)掺入量和三磷酸肌醇[Ins(1,4,5)P3]累积量。结果:U46619在一定范围内(1 nmol/L~100 nmol/L)以浓度依赖的方式增加人气管平滑肌的细胞数(P<0.01)。U46619也增加[3H]-TdR的掺入量和Ins(1,4,5)P3的累积量(P<0.01)。磷脂酶C抑制剂新霉素阻止Ins(1,4,5)P3累积量的增加(P<0.01),但对U46619的有丝分裂原的活性没有作用。结论:显示U46619刺激培养的人气管平滑肌细胞增殖。 主题词 肌,平滑;增生;哮喘
Role of U46619 for proliferation of cultured human airway smooth muscle cell

Abstract AIM:To study the role of U46619 in stimulating the proliferation of cultured human airway smooth muscle cell (ASMC).METHODS:Human ASMC were isolated and subcultured, varying concentrations of U46619 were added to the media. Cell counts were obtained,[3H] -thymidine incorporation ([3H]-TdR) and inosital 1,4,5-trisphosphate [Ins(1,4,5)P3] accumulation were measured.RESULTS:U46619(1 nmol/L-100 nmol/L) increased cell number in a concentration depend manner (P<0.01) U46619 also increased [3 H]-TdR and (1,4,5)P3 accumulation (P<0.01). The latter response was blocked by phospholipase C inhibition with neomycin (P<0.01),however, neomycin had no effect on the promitogenic action of U46619.CONCLUSION: U46619 stimulates the proliferation of cultured human ASMC. 血栓素A2(thromboxane A2,TXA2)是花生四烯酸经环氧化酶途径代谢的主要产物之一,能强烈地收缩血管和气管平滑肌,其收缩血管的作用比血管紧张素Ⅱ强100倍[1]。TXA2性质不稳定,半衰期短。近来的研究显示:气道平滑肌的增生是引起哮喘患者气道狭窄的重要因素[2]。许多炎性介质如组胺、内皮素-1、白三烯D4等均能引起气管平滑肌增殖[3,4],已有报道TXA2引起兔气管平滑肌增殖[5]。我们通过离体培养的人气管平滑肌细胞,观察TXA2稳定的类似物U46619对其增殖的作用。
材料与方法
