牛轮状病毒受体分析 |

【摘要】目的 分析牛轮状病毒受体的理化性质及该受体在气管粘膜组织的分布。方法 采用SDS-聚丙烯酰胺凝胶电泳法(SDS-PAGE)、酶消化法、固相分析法对纯化的牛轮状病毒受体进行分析,以免疫组化法检测了新生牛气管粘膜细胞表面牛轮状病毒受体。结果 牛轮状病毒受体SDS-PAGE分子量为48000,该受体对胰蛋白酶及Pronase E 蛋白酶敏感,经Clostridium perfringens及Arthrobacter ureafaciens神经氨酸酶作用后,受体失去结合病毒的生物学活性。免疫组化试验结果为阳性。结论 实验结果表明牛轮状病毒受体是结构复杂的糖蛋白,唾液酸残基是受体与牛轮状病毒作用位点的组成部分。新生牛气管粘膜细胞表面有牛轮状病毒受体存在。 【主题词】 轮状病毒 受体,病毒 腹泻
Analysis of the receptor for bovine rotavirus Li Jinhui,Zi Ziqiang,Ren Zhongyuan. Department of Microbiology,Tianjin Medical University,Tianjin 300070 【Abstract】 Objective We studied the physical and chemical characteristics of the receptor for bovine rotavirus and the distribution of the receptor on the tracheal mucous membrane. Methods The methods of SDS-polyacrylamide gel electrophoresis,enzyme digestion assay and solid-phase assay were used to analyze the purified receptor for bovine rotavirus.The receptor on the cell surface of the tracheal mucous membrane of neonatal bovine was verified by immunohistchemistry method. Results The receptor molecular weight was 48kD on SDS-polyacrylamide gel electrophoresis.It was sensitive to trypsin and Pronase E.Treated by the neuraminidase from Clostridium perfringens or Arthrobacter ureafaciens,the receptor lost its biological activity of binding virus.The assay result of immunochemistry was positive. Conclusions All these results above showed that the receptor for bovine rotavirus was a kind of glycoprotein with a complex structure.Sialic acid remnant is infected to be a main part of the receptor site reacting with bovine rotavirus.The receptor for bovine rotavirus has existed on the cell surface of the tracheal mucous membrane of neonatal bovine. 【Subject words】 Rotavirus Receptor Diarrnea
轮状病毒(rotavirus,RV)是婴幼儿腹泻的最重要病原体,但其致病机制至今仍不清楚,亦无十分有效的防治方法。病毒感染的第一步是与宿主细胞表面受体的吸附,是病毒感染的关键。目前有关RV受体研究的报道几乎均局限于病毒颗粒与细胞相互作用特性的研究,不能完全排除细胞其它成份的影响〔1〕。为此,本实验对分离纯化的牛轮状病毒(bovine rotavirus,BRV)受体进行研究,精确分析其结构及功能。
