藻酸双酯钠与细胞因子对巨核细胞生成的调控作用

  摘 要 目的和方法:采用血浆凝块(PC)、无血清琼脂(SFA)培养及[125 I]-bFGF 结合试验等方法研究PSS及TSP等8种细胞因子对巨核细胞生成的作用与机制。结果:在PC系统,PSS刺激CFU-MK形成;在SFA系统,PSS无此作用。PSS加强bFGF或再障血清对CFU-MK 形成的刺激作用,但PSS不增加bFGF与其高亲和力受体的结合。TSP及具有肝素结合位点(HBD)的TSP多肽片段明显抑制CFU-MK的形成,无HBD的TSP片段则无此作用。TSP、PF4及TGFβ1对巨核细胞的抑制作用可被PSS所逆转。结论: PSS对巨核细胞生成起正调控作用,其机制与增强巨核细胞生成刺激因子活性及中和抑制因子的活性有关;TSP是巨核细胞生成负调控因子,HBD是其重要功能基团。

Modulating effect of PSS and cytokines on megakaryocytopoiesis in vitro

  Abstract AIM and METHODS: The effect of propylene glycol alginate sodium sulphate (PSS), eight cytokines including thrombospondin (TSP), PF4, TGFβ1, IL-3,IL-6, EPO, GM-CSF, bFGF and aplastic pig serum (APS) known to be rich in TPO, alone or in combination, on megakaryocytopoiesis and the mechanism of PSS action were studied by plasma clot system (PCS), serum free agar system (SFAS) and [125I]-bFGF binding test. RESULTS: PSS stimulated the growth of CFU -MK in PCS, but not in SFAS and showed no effect on the growth of CFU- GM in PCS and SFAS. PSS, combined with bFGF or APS, resulted in an additive stimulation on the growth of CFU-MK and it did not enhance the binding of bFGF to its high affinity receptor. TSP and TSP fragment comprising residues 1-174(TSP1-174) with heparin-binding domain (HBD), but not the TSP559-669 without HBD obviously inhibited the growth of CFU-MK from unfractionated or fractionated Lin- marrow cells. The inhibitory effect of TSP, PF4 or TGFβ1 on the growth of CFU-MK could be abrogated by PSS. CONCLUSION: (1) PSS is a positive modulator of megakaryocytopoiesis by a mechanism of intensifing megakaryocyte colony-stimulating factors such as bFGF, TPO, etc. and neutralizing the inhibitory factors such as TSP, PF4, TGFβ1, etc. and (2) TSP is a negative modulator of megakaryocytopoi
