摘要 目的:初步探讨L-精氨酸对高胆固醇血症鹌鹑主动脉粥样斑块面积的影响。方法:选择健康雄性日本鹌鹑80只,分为正常对照组、高脂组、L-Arg组和安慰剂组。前8周高脂饮食,复制动脉粥样硬化模型。在此基础上,后8周L-Arg组与安慰剂组停喂胆固醇和油脂,饮水补充2.25%L-精氨酸,后者饮自来水。在8周末、16周末,取出主动脉,形态学观察动脉粥样硬化病变;计算斑块面积百分比;收集血清作生化分析。结果:(1)高脂组斑块面积从8周末的37%上升到16周末50%,内膜从40 μm增厚到56 μm;(2)L-Arg组主动脉斑块面积和内膜厚度明显减少;(3)补充2.25%L-精氨酸8周,血清总胆固醇(TC)和丙二醛(MDA)水平恢复正常,超氧化物歧化酶(SOD)活性比高脂组高80%,比安慰剂组高60%。结论:L-Arg可能通过降血脂,减少脂质过氧化损伤而具有明显缩小实验性鹌鹑主动脉粥样硬化斑块面积的作用。 主题词 精氨酸; 动脉粥样硬化; 胆固醇
Effects of L-arginine on atherosclerotic plaque area of aortae of quails with hypercholesterolemia

Abstract AIM:To study the effect of supplemental L-arginine on preexisting atherosclerotic lesions.METHODS: Male Japanese Quails received normal chow (control group) or 1% cholesterol and 3% fat chow (high lipid group) for 8 weeks. Subsequently, L-arginine (2.25% in drinking water; L-Arg group) or Vehicle (vehicle group) was administered for an additional 8 weeks, while the high cholesterol and fat was cancelled. At weeks 8, 16, aortae were harvested for histomorphological study as well as plaque surface area and intimal thickness determination. Serum was obtained for biochemical analysis. RESULTS:Planimetry of the luminal surface and intimal thickness of the aortae from high lipid group revealed a progressive increase. Animals in the L-Arg group manifested improved activity of superoxide dismutase (SOD) and decreased level of total cholesterol (TC) and malondialdehyde (MDA) in serum and with a reduction of plague area and intimal thickness at 16 weeks. CONCLUSION:Hypercholesterolemia induces progressive plaque lesion and intim hyperplasia. L-arginine might induce regression of preexisting atherosclerotic lesions, and involve in its lowering lipids and decreasing lipid peroxide injury. MeSH Arginine; Atherosclerosis; Cholesterol
