血管内皮生长因子及其基因在自发性高血压大鼠心肌中的表达 |

摘 要 目的:血管内皮生长因子(VEGF)是新近确定的一 种特异作用于血管内皮细胞的活性肽。最近发现正常心肌细胞有VEGF及其基因表达,但对高 血压肥大心脏心肌VEGF及其基因表达的变化尚不清楚。方法:本实验采用 免疫组化和分子杂交方法,对自发性高血压大鼠(SHR)肥大心脏心肌VEGF及其基因表达进行 研究。结果:免疫组化结果表明,SHR心肌细胞浆内特异性VEGF染色颗粒明 显多于WKY对照大鼠。Northern分子杂交结果表明,SHR心肌VEGF mRNA表达明显强于WKY对照 大 鼠。结论:目前对这一结果的生理意义还不清楚,推测可能与心肌肥大时 细胞间质血管增生有关。 主题词 生长物质;内皮,血管;心肌;肥大;高血压
Expression of vascular endothelial growth factor and its gene in myocardium of spontaneously hypertensive rats

Abstract AIM:Vascular endothelial growth factor(VEGF), a growt h and permeability factor specific for endothelial cell, has been identified in m yocardium of the normal adult heart, but the change of VEGF in myocardium of hyp ertensive hypertrophic heart was unclear. METHODS:We therefor stu d ied the expression of VEGF and its gene in myocardium of spontaneously hypertens ive rats(SHR) by immunohistochemistry assay and Northen bolt. The results were analyzed by scanning densitometry.RESULTS:Compared with control W istar Kyoto (WKY) rats, the left ventricle weight/body weight of SHR increased b y 61%(P<0.01). The intracellular VEGF immunoactivity was present in myocard ium of WKY rats and SHR, but the immunoactivity of SHR was 91% strong er than that of WKY rats. The results of Northern bolt showed that the expressio n of VEGF mRNA of SHR increased by 67% than that of WKY by scanning densitometr y. CONCLUSION:Although little is known about the role of expressi on upregulation of VEGF and its gene in myocardium of SHR, it seems to respond to vascular proliferation of hypertrophic heart.
