黄芪对感染病毒大鼠心肌细胞钙通道及钠钙交换载体的效应 |

摘 要 目的与方法:以柯萨奇B3病毒感染急性分离的 成年大 鼠心肌细胞,利用膜片钳记录技术了解黄芪对感染柯萨奇病毒后心肌细胞L型钙通道及钠钙 交换载体的影响,以探讨黄芪用于心肌炎治疗的作用机制。结果:黄芪不 影响正常心肌细胞L型钙通道电流的幅度,但可减缓L型钙通道电流的衰减。黄芪也不影响内 向NCE电流,但使外向NCE电流增加,从而使动作电位期间由NCE转运入细胞内的钙离子增加 。病毒感染增加心肌细胞L型钙通道电流的幅度,抑制内向钠钙交换电流,并使钠钙交换电 流的逆转电位负移。黄芪可抑制病毒感染细胞L型钙通道电流的增加, 减轻CVB3对内向NCE电流的抑制作用和 使负移的逆转电位正常化。结论:黄芪通过拮抗CVB3感染对L型钙通道 和钠钙交换电流的影响,可防止病毒感染可能导致的细胞内钙超载和异常电活动,从而对感 染细胞起到保护作用。 主题词 黄芪;柯萨奇病毒;钙通道;心肌;大鼠;

Abstract AIM and METHODS: By patch clamp recording techniques to investigate the effects of astragalus membranaceus (AM) on L type voltage dep endent calcium channel (LVDCC) and Na+/Ca2+ exchange (NCE) in a dult ra t cardiomyocytes infected with coxsackievirus B3(CVB3) so as to understand the mechanism of AM to cure myocarditis.RESULTS:AM did n ot affect the amplitude of calcium current,but decreased the “run down”of calc ium current;also,AM had no effect on inward NCE current,but increased outward NC E current.Both of the functions of AM to LVDCC and NCE would increase the calciu m inflow during the action potential.
