中药海乐得对部分肾切除大鼠残余肾脏表达PAI |

摘 要 目的：观察中药制剂海乐得对5/6肾切 除(5/6NX) 大鼠残余肾组织表达纤溶酶原激活物抑制物-1(Plasminogen activator inhibitor-l， PAI -l) 、转化生长因子β(transforming growth factor-β，TGFβ)的影响。方法：海乐得治疗5/6肾切除大鼠6周后处死，检测尿蛋白排泄和血生化变化，制作残肾组织病 理和冰冻切片，提取残余肾脏组织RNA，分别采用组织原位杂交及Northern印记杂交方法观 察PAI-l、TGFβ的基因表达。结果：中药治疗组残肾组织中PAI-l和TGFβ mRNA表达、尿蛋白排泄、血清 尿素氮和肌酐均明显低于未治疗组，肾脏病理损害以及脂代谢紊乱亦有明显改善。 结论：海乐得通过PAI-l和TGFβ途径，能明显改善慢性肾功能衰竭大鼠的肾功能和 病理损伤。

Effects of HaiLeDe on mRNA expressions of PAI-l and TGFβ
in the rat remnant kidney

Abstract AIM：To explore effects of Traditional Chinese Medic in e(TCM) prescription HaiLeDe on the mRNA expression of plasminogen activator inh ibitor-l(PAI-l) and transforming growth factor-β(TGFβ) in the remnant kidney o f Wistar rats with 5/6 nephrectomy.METHODS： The Wistar rats with 5 /6 nephrectomy were administered HaiLeDe for six weeks and then sacrificed. Prot einuria and serum biochemical parameters were examined. Expressions of PAI-l and TGFβ mRNAs in the remnant kidney of the rats with 5/6 nephrectomy were determined by in situ hybridization and Northern blot hybridization analy sis. RESULTS：The mRNA expressions of PAI-l and TGFβ genes were signif icantly increased in the renal tissue of 5/6 nephrectomized rats and the treatme nt with HaiLeDe markedly decreased the mRNA expressions of PAI-l and TGFβ genes. P roteinuria, blood urea nitrogen, serum creatinine, triglyceride and glom erular hypertrophy and sclerosis were also redu ced significantly in the 5/6 nephrectomized rats treated with HaiLeDe compared w ith those in the non-treated group.
