沙土鼠及大鼠局部脑缺血时血小板活化因子的变化 |

摘 要 目的:血小板活化因子(PAF)拮抗剂能改善局部脑缺血所致功能和组织损伤,本文在沙土鼠单侧脑缺血及大鼠大脑中动脉凝闭的局部脑缺血观察了缺血后大脑组织PAF的变化。方法:PAF的测定采用薄层层析进行PAF分离提取,用生物法测定PAF含量的变化。结果:1.沙土鼠右侧颈总动脉结扎3 h再灌2 h,右侧大脑组织PAF含量明显高于左侧。2.大鼠左侧脑中动脉凝闭后48 h,左侧大脑组织PAF含量明显高于右侧。结论:PAF在局部脑缺血继发神经元损伤中可能有一定作用。 主题词 血小板活化因子;局部脑缺血;沙鼠亚科;大鼠
Platelet-activating factor in brain after regional ischemia in gerbils and rats

Abstract AIM:Platelet-activating factor(PAF) bas been reported to be an active mediator of neuronal damage in regional cerebral ischemia on the basis of indirect pharmacological data from PAF antagonists. The direct measurement of PAF has not been reported previously in regional cerebral ischemia. It prompted us to investigate the change of PAF in unilateral ischemia in gerbils and regional ischemia in rats.METHODS:Thin layer chromatography was combined with biological methods for the determination of PAF in brain after cerebral ischemia.RESULTS:(1) The PAF content of ischemic brain was increaesd significantly in gerbils subjected to 3 h unilateral cerebral ischemia followed by 2h reperfusion respectively(4.40±0.90) and (1.30±0.40) pg/g, P<0.05. (2)The PAF content of ischemic brain was increased significantly also in rats subjected to 48 h regional ischemia respectively(5.74±0.75) and (0.76±0.05)pg/g, P<0.05. CONCLUSION:PAF could contribute to secondary neuronal damage after regional cerebral ischemia. MeSH Platelet activating factor; Cerebral ischemia; Gerbillinae; Rats
血小板活化因子(platelet-activating factor, PAF)为一很强的非特异性磷脂介质,参与多种病理过程。PAF可诱导血小板聚集,引起脑血管收缩,减少大脑局部血流量,增加血脑屏障通透性,激活中性白细胞,形成脂质过氧化物[1]。近年对PAF在脑缺血继发神经元损伤中的作用,引起了关注[2]。但关于局部脑缺血时大脑组织PAF的变化,文献中尚未见报道,PAF在局部脑缺血发病中的作用系来自间接证明,即应用PAF拮抗剂可减轻局部脑缺血后机能和组织损伤[3]。本实验目的在于观察沙土鼠单侧脑缺血及大鼠局部脑缺血时大脑组织PAF含量的变化,进一步证明PAF在局部脑缺血继发神经元损伤发病中的作用。
