内皮素抗体在大鼠肾缺血 再灌注损伤中的作用 |

摘 要 目的和方法:为探讨内皮素在肾缺血/再灌注损伤中的发病学意义,本实验在大鼠肾缺血/再灌注损伤模型上,观察了内皮素抗体对肾缺血/再灌注损伤的作用。结果:肾缺血/再灌注损伤大鼠,血清尿素氮(SUN)、肌酐(SCr)含量及血浆内皮素(PET)水平显著升高;脂质过氧化物丙二醛(MDA)产生增多;肾小管上皮细胞明显损伤。应用内皮素单克隆抗体能显著降低PET水平和MDA含量,减轻肾小管上皮细胞损伤,改善肾功能。结论:内皮素是参与肾缺血/再灌注损伤发病的重要因素之一,阻断ET的生物学作用是防治肾缺血/再灌注损伤的有效途径。 主题词 肾;局部缺血;再灌注;内皮缩血管肽类;抗体
The role of ET-Ab in renal ischemia-reperfusion injury in rats

Abstract AIM and METHOD In order to evaluate the possible role of ET-Ab in the pathogenesis of renal ischemia-reperfusion injury, the effect of ET-Ab on renal ischemia-reperfusion injury was investigated on a model produced by bilateral clamping the renal artery and vein followed by reperfusion in rats.RESULTS:Contents of SUN, SCr, MDA and level of plasma endothelin (PET) were significantly increased in renal ischemia-reperfusion injured rats, and the impairment of renal tubular epithelial cells was observed. ET-Ab might siginificantly decreased contents of SUN, SCr, MDA and level of PET, and attenuate the renal ischemia-reperfusion injury of rats.CONCLUSION:ET plays an important role in the pathogenesis of ischemia-reperfusion injury. Blocking the biological actions of ET might be an effective means of preventing and treating ischemia-reperfusion injury. MeSH Kidney; Ischemia, Reperfusion; Endothelins; Antibodies
内皮素(endothelin,ET)是血管内皮细胞分泌和释放的一种血管活性多肽,是迄今已知体内作用最强的缩血管物质[1],研究表明,缺血性急性肾功能衰竭时,血浆内皮素水平明显升高,ET受体上调[2,3]。但也有研究认为,缺血性急性肾功能衰竭时肾小球ET受体无变化[4]。目前,有关ET在肾缺血/再灌注损伤中的作用及其机制尚不十分清楚。本工作应用内皮素单克隆抗体(ET-Ab),观察其对大鼠肾缺血-再灌注损伤的作用,以进一步验证ET在肾缺血/再灌注损伤中的发病学意义,并探索有效的防治措施。
材料与方法
