内皮素在自发性高血压大鼠肾内小动脉重建中的作用 |

【摘要】 目的 探讨内皮素在高血压肾内小动脉重建中的作用。 方法 4周龄SHR,给予口服型内皮素A型受体拮抗剂BMS-182874至16周,在肾组织切片上分别用光镜和电镜配合计算机图像分析法观测肾内小动脉的几何形态学指标和小动脉平滑肌及其间隙,离体肾脏灌流法测定最小肾血管阻力。 结果 BMS-182874在没有降低SHR血压的情况下,使外径在100~200μm之间的小动脉管壁厚、壁面积、壁厚内径比和SMC宽度都显著减小。 结论 内源性内皮素在一定程度上促进了SHR肾内小动脉的重建。 【关键词】 小动脉;肾;内皮素;受体拮抗剂;重建;自发性高血压大鼠 【中图分类号】R544.102 【文献标识码】A 【文章编号】 0529-1356(2000)02-137
THE ROLE OF ENDOTHELINE ON THE REMODELING OF THE RENAL ARTERIOLES IN SPONTANEOUSLY HYPERTENSIVE RATS

【Abstract】 Objective To investigate the role of endothelins on remodeling of renal arterioles in hypertension. Methods ET-A receptor antagonist BMS-182874 was given orally to 4 week-old spontaneously hypertensive rats(SHR) for 12 weeks. The geographic parameters of renal arterioles, and the widths of vascular smooth muscle cells(SMC) and intercellular space were studied on kidney slices respectively by light microscope and electromicroscope combining computer-assistant image analysis system. The minimal renal vascular resistance was studied by isolated kidney perfusion system. Results BMS-182874 decreases markedly the wall thickness, wall area, ratio of wall thickness to inner diameter and width of SMC of renal arterioles in 100 to 200μm outer diameter. Conclusion Endogenous endothelins take part in remodeling of renal arterioles in SHR in some extent. 【Key words】 Arteriole; Renal;Endothelins; Receptor antagonist; Remodeling;Spontan eously hypertensive rats
