摘 要 目的和方法:在培养的家兔血管平滑肌细胞上,观察了肾上腺髓质素(Adm)对溶血磷脂酸(LPA)诱导的DNA合成和丝裂素活化蛋白激酶(MAPK)活性途径激活的影响。结果:LPA使[3H]-TdR掺入和MAPK活性增加,Adm对基础水平的MAPK活性和[3H]-TdR的掺入并无显著影响,但Adm对LPA刺激[3H]-TdR掺入和MAPK活性具有抑制作用。结论:Adm和LPA相互作用参与血管平滑肌细胞增殖的调节。 主题词 肌,平滑,血管;细胞;家兔
Adrenomedullin inhibits the lysophosphatidic acid-induced proliferation of vascular smooth muscle cells

Abstract AIM and METHODS:The effect of adenomedullin(Adm) on lysophosphatidic acid(LPA)induced DNA synthesis and mitogen-activated protein kinase(MAPK) activity in cultured vascular smooth muscle cells(VSMCs) of rabbit was observed.RESULTS:LPA enhanced the cultured VSMCs [3H]-TdR incorporation and increased MAPK activity concurrently. Adm could not affect basal MAPK activity or [3H]-TdR incorportion,but inhibit LPA-induced [3H]-TdR incorporation and MAPK activity in a concentration-dependent manner.CONCLUSION:The action between Adm and LPA may play an important role in regulating the proliferation of VSMCs. 血管平滑肌细胞(vascular smooth muscle cells,VSMCs)增殖在高血压、动脉粥样硬化和血管形成术后再狭窄等心血管疾病中具有重要发病学意义,确切增殖机理目前尚未完全阐明,已知受到体内多种神经-内分泌因子的网络调节所控制[1]。溶血磷脂酸(lysophosphatidic acid,LPA)是近年新发现的磷脂类信号分子,心血管组织富含其受体,本室实验表明LPA是VSMC的强效丝裂素原,促增殖作用与激活细胞内丝裂素活化蛋白激酶(mitogen-activated protein kinase,MAPK)途径有关。肾上腺髓质素(adrenomedullin,Adm)是强效舒血管肽,具有抑制血清、生长因子和内皮素等激活MAPK和促进平滑肌细胞增殖的作用[2]。本工作在培养的家兔血管平滑肌细胞上,观察Adm对LPA诱导的DNA合成和MAPK途径激活的影响,以认识Adm与LPA对VSMC增殖调节的相互作用。
材料与方法
