牛磺酸对大鼠脑缺血再灌注损伤时细胞凋亡的影响 |

摘 要 目的:研究牛磺酸(taurine, Tau)对脑缺血再灌注损伤中细胞凋亡的影响。方法:建立大鼠全脑缺血再灌注模型,使用DNA片段原位末端标记(terminal deoxynucleotigyl transferase-mediated dUTP nick end labeling, TUNEL)和流式细胞仪(flow cytometry, FCM)观察了Tau治疗后细胞凋亡的变化。结果:海马及顶叶皮层内凋亡细胞数显著降低,但在1 mg及2 mg剂量组间无显著差异。结论:Tau对脑缺血再灌注损伤中神经细胞的保护作用可能部分由于降低细胞凋亡的发生。 主题词 脑;局部缺血;牛磺酸
The effect of taurine on apoptosis during ischemia-reperfusion injury in rats

Abstract AIM and METHOD:In order to recognize the effect of taurine on apoptosis during ischemia-reperfusion injury, the model of rat globle brain ischemia-reperfusion injury was made, the changes of apoptotic cells were observed after taurine treatment by using methods of terminal deoxynucleotigyl transferase-mediated dUTP nick end labeling (TUNEL) and flow cytometry(FCM).RESULTS:The number of apoptotic cells decreased significantly in hippocampus and parietal cortex, but there were significant difference between 1 mg and 2 mg dose group. CONCLUSION:The neural protective effect of taurine against ischemia-reperfusion injury may be partially due to reducing occurrence of apoptosis. MeSH Brain; Ischemia; Taurine
近年的研究提示细胞凋亡是脑缺血损伤的重要形式[1]。牛磺酸(taurine, Tau)是人体内含量较高的自由氨基酸,在中枢神经系统中具有重要生理功能,是一种抑制性作用的氨基酸[2]。研究发现Tau对缺血、惊厥等引起的神经元损害具有对抗作用[2,3],这种抗损害作用是通过对抗兴奋性氨基酸、过氧化物和调节细胞内Ca2+水平实现的,而兴奋性氨基酸、过氧化物和细胞内Ca2+升高与细胞凋亡(apoptosis)的发生有关[4,5]。因此,Tau可能对缺血后细胞凋亡有一定影响。本研究通过建立全脑缺血模型,观察Tau治疗后细胞凋亡的变化,为脑缺血的防治提供实验依据。
材料与方法
1.大鼠脑缺血动物模型:Wistar大鼠,体重250~300 g,0.3%戊巴比妥钠(ip)麻醉后,固定于立体定位仪上,使用四血管阻塞模型,先在第1、2颈椎水平闭合双侧椎动脉,分离双侧颈总动脉。次日在乙醚麻醉下,用微动脉夹夹闭双侧颈总动脉,缺血30 min,松开微动脉夹,进行再灌流,达到再灌流时间后取材。 2.牛磺酸给药方式及分组:采用脑室内给药,将大鼠固定于立体定位仪,颅顶保持在同一水平面,以前囟为零点,向后0.8 mm,外侧1.5 mm,深4.5 mm,进入脑室(右)后推药时感阻力小。以1 mg及2 mg两个剂量组,分别给以10%牛磺酸([1] [2] [3] 下一页 上一个医学论文: 异丙肾上腺素致大鼠心肌坏死时心肌细胞核钙转运功能的变化 下一个医学论文: NF
