【摘要】 目的 观察胶质细胞源性神经营养因子受体-α(GDNFR-α)在体外培养的大鼠脊髓和背根节神经元中的分布,探讨GDNF对脊髓运动神经元和感觉神经元的作用。 方法 原代培养脊髓和背根节神经元,5d后行抗GDNFR-α多克隆抗体免疫组织化学SP法染色。 结果 GDNF免疫反应存在于体外培养的脊髓神经元、背根节神经元以及胶质细胞中。 结论 GDNF可能对脊髓神经元、背根节神经元和胶质细胞的生理功能具有一定的调节作用。 【关键词】 胶质细胞源性神经营养因子受体-α;脊髓;背根节;大鼠 【中图分类号】 R322.81 【文献标识码】 A 【文章编号】 0529-1356(2000)03-215

【Abstract】 Objective In order to investigate the effect of glial cell line-derived nerurothophic factor(GDNF)on the neurons of spinal cord and dorsal root ganlia(DRG),we observed the localization of GDNFR-α on neurons of spinal cord and DRG of rat in vitro. Methods The neurons of spinal cord and DRG of rats were cultured.Five days later,these cells were stained with GDNFR-α polyclone immunohistochemical staining technique. Results The positive GDNFR-α immunoreactivity was present in the neurons of spinal cord and DRG and glial cells. Conclusion The present results suggested that GDNF might have some important effects on the neurons of spinal cord and DRG and glial cells in vitro. 【Key words】 Glial cell line-derived neurothophic factor receptor-α(GDNFR-α); Spinal cord; Dorsal root ganglia(DRG);Rat
胶质细胞源性神经营养因子(glial cell line-derived neurotrophic factor,GDNF)是在1993年由小鼠胶质细胞系B49中分离出的糖基化的二硫键结合的同源二聚体蛋白质,属于转化生长因子β(transforming growth factor-β)超家族的成员[1]。研究发现GDNF是一种多效能的神经营养因子。不仅对中脑多巴胺能神经元有特异性作用,促进多巴胺能神经元的存活、生长、多巴胺的摄取、再生和分化,而且对神经系统运动神经元和感觉神经元的发育、存活和再生有着巨大的潜能[2~4]。GDNF的受体为多成分的复合物,是由固定于质膜外层的糖基磷脂酰肌醇(GPI)被称为GDNFR-α和GDNF功能性[1] [2] [3] 下一页 上一个医学论文: 大鼠肝内起始淋巴管超微结构观察 下一个医学论文: 突触体素 S
