散发性戊型肝炎病毒部分核苷酸序列分析 |

【 摘要 】 目的 为阐明戊型肝炎病毒(HEV)毒株的地理分布、流行特征以及研制血清学 诊断试剂和基因工程疫苗提供资料。方法 选择1例浙江仙居山区散发性HE患者,从其血清 中分离HEV RNA,通过逆转录-套式聚合酶链反应法(RT-nPCR),扩增该散发性HEV(Z-33株) ORF2区部分cDNA片段(497bp),然后进行直接序列分析,并与HEV各主要代表株序列作比 较。结果 Z-33株与新疆流行株CH1.1的核苷酸及氨基酸序列同源性分别为96.7%及98. 2%;与缅甸流行株的同源性分别为94.2%及99.1%;与缅甸散发株的同源性分别为95.2% 及99.1%,与墨西哥株的同源性分别为81.8%及94.5%。 结论 浙江仙居散发性HEV Z-33 株和新疆流行株CH1.1一样, 与HEV缅甸株可能为同一亚型。

【 Abstract 】 HEV RNA was extracted from the serum of a patient with s poradic hepatitis E in Xianju, Zhejiang, designated as HEV Z-33 strain. 497 bases of HEV cDNA in ORF2 region was amplified by a reverse transcrip tion nested polymerase chain reaction(RT-nPCR),sequenced directly an d compared with those of other HEV strains.The results demonstrated th at nucleotides and amino acids of the HEV Z-33 were 96.7% and 98.2% homolo gous with those of the Xinjiang epidemic strain of HEV CH 1.1; 94.2% and 99.1% with the Burma epidemic strain of HEV(B);95.2% and 99.1% with the B urma sporadic strain of HEV(B);81.8% and 94.5% with the Mexico strain of HEV(M), respectively. It suggests that the HEV Z-33 strain may belong to t he HEV(B) subtype, as the CH1.1 strain does.
