苯丙氨酸对自发性高血压大鼠血管平滑肌细胞内钙的影响

  摘 要 目的:探索苯丙氨酸对自发性高血压大鼠血管平滑肌细胞内钙的影响。方法:将SHR和WKY的血管平滑肌细胞在不同浓度苯丙氨酸培养液中培养一定时间后,加入[45CaCl2]温育一定时间后,测定进入细胞内[45Ca2+]的量;而后将在不同浓度苯丙氨酸培养液中培养一定时间后的SHR和WKY的血管平滑肌细胞同PSS液温育,采用Fura-2/AM荧光指示剂测定不同浓度苯丙氨酸对细胞内游离Ca2+的影响。结果:苯丙氨酸浓度越高,进入SHR血管平滑肌细胞内的钙和细胞内游离钙含量越少,但对WKY无影响。结论:苯丙氨酸可以特异性地抑制Ca2+进入SHR血管平滑肌细胞和使细胞内游离Ca2+减少。

L-phenylalanine effects on intracellular calcium in
vascular smooth muscle cell of spontaneously hypertensive rat

  Abstract AIM:To find the L-Phenylalanine (Phe) effects on intracellular calcium in vascular smooth muscle cell(VSMC) of spontaneously hypertensive rat (SHR) and WKY.METHODS:VSMC of SHR and WKY were incubated with phenylalanine in 7 different concentration for a period, and then added[45CaCl2] into each well and incubated for another 30 minutes. The VSMC in each well were washed and counts.min-1 of[45CaCl2] was measured. Similar to above, incubated VSMC cultured in different concentration Phe with PSS solution, and measured the intracellular free calcium concentration with Fura-2/AM. RESULTS:The higher L-phenylalanine concentration, the lesser calcium influx from the extracellular and intracellular free calcium concentration in VSMC of SHR. However, there were no variations in VSMC of WKY. CONCLUSION:Phenylalanine effect that reduces total intracellular free calcium concentration may contribute to its action in preventing blood pressure rise and the hypertrophy of vessel in SHR.

  血管平滑肌细胞(vascular smooth muscle cell, VSMC)的生长加速、增殖和收缩是高血压血管平滑肌的特征性表现,也是高血压发病的主要原因[1]。生长因子、激素和自体效应物质促使细胞的有丝分裂[2]。钙作为重要的细胞内信使是各种促生长因素最终发挥其促细胞增殖作用的重要环节[3],是多种生长活性肽分子发挥其促增殖作用的共同途径。钙拮抗剂可以抑制VSMC的增生和迁移[4,5]。我们的研究曾表明,L-苯丙氨酸可以预防双亲高血压病患者子女的血管壁增厚。本研究以自发性高血压
