摘 要 目的：研究凋亡基因加强表达对鼻咽癌细胞株程序性死亡的影响。方法：用脂质体转染法将已构建的pSFFV-baxα-neo质粒导入CNE2细胞中，RT-PCR， 间接免疫荧光检测转染后CNE2中baxα表达水平；流式细胞术及电泳检测凋亡。结果：脂质体转染质粒pSFFV-baxα-neo入CNE2细胞，RT-PCR随机检测6个阳性克隆中baxα的mRNA表达水平明显提高，间接免疫荧光检测CNE2-baxα中Baxα强阳性(高)、中强阳性(中)以及弱阳性(低)表达。流式细胞仪分析细胞凋亡率显示高、中表达Baxα的细胞株凋亡率为11.3%、7.9%，显著高于对照株5.8%的凋亡率(P＜0.001)，低表达Baxα的细胞株凋亡率为6.1%，与对照株无明显差异(P＞0.05)。结论：Baxα在CNE2中促进无血清诱导的细胞凋亡。

Overexpression of the death-promoting gene baxα enhances apoptosis of nasopharyngeal carcinoma cell line CNE2

Abstract AIM:To investigate the effect of baxα overexpression on nasopharyngeal carcinoma (NPC) cell line CNE2 in vitro. METHODS:Plasmids pSFFV-baxα-neo were transfected into CNE2 cell in liposome. RESULTS：Levels of baxα were very high in six clones randomly which were assayed with RT-PCR.Strong positive (high), medium-strong positive (medium) and weak-positive (low) Baxα expression in the CNE2-baxα cell line by indirect immunoflourescence staining. After serum deprivation for 48 h,the apoptotic rates of high, medium expression of baxα were 11.3%, 7.9% respectiviely, significantly higher than that of control cell line transfected with empty vector 5.8% (P＜0.001). The apoptotic rate of the cell line with low Baxα expression is 6.1%. There was less significant difference compared with control cell line (P＞0.05).CONCLUSION：Baxα enhances the serum-deprivation induced apoptosis in CNE2 cell line.
