热休克蛋白70与家兔颈总动脉球囊损伤后增殖细胞核抗原表达的关系 |

摘要 目的:研究热休克蛋白70(HSP70)与经皮冠状动脉腔内成形术后再狭窄形成中细胞增生的关系。方法:复制家兔颈总动脉球囊损伤模型,用免疫组化方法检测术后3、9、14、28 d损伤动脉壁中HSP70和增殖细胞核抗原(PCNA)表达水平;用计算机图像分析系统定量,并对两者进行相关分析。结果:损伤动脉壁HSP70及PCNA检测呈阳性,正常动脉呈阴性。9 d时两者表达指数均较各自3 d时显著升高,14 d均达峰值,28 d显著降至3 d时水平。时相相关分析两者显著正相关。结论:细胞增生活性升高伴随HSP70的产生增多;动脉损伤时机体可能通过HSP70合成增多而促进细胞再生,一定程度上促成再狭窄。 主题词 热休克蛋白质类;抗原;动脉
The relationship between expression of HSP70 and PCNA in ballooned rabbit carotid

Abstract AIM:To investigate the expression of heat shock protein 70 (HSP70) involving in the development of restenosis after percutaneous transluminal coronary angioplasty (PTCA).METHOD:The rabbit carotid balloon injury model was used.Six animals were killed at 3 days, six at 9 days, six at 14 days and six at 28 days after injury respectively, and the ballooned carotid segments were removed.Then, immunohistochemistry combined with computer image analysis was used to determine HSP70-expression index (HEI) and proliferating cell nuclear antigen (PCNA)-expression index (PEI) in injured arteries.RESULTS:Both stainnings were negative in normal vessels while positive in injured vessels.HEI and PEI significantly increased at day 9 compared with day 3 and reached their maxiums at day 14, then declined at day 28 significantly.The time course of cell proliferation was significantly positive correlated with that of the HSP70-expression.CONCLUSION:Cell proliferation is accompanied by production of HSP70.It seems that body may induce cell proliferation by increasing the synthesis of HSP70, thus to accelerate the repairment of injured artery.
