人红细胞老化过程中花生凝集素和Con A受体电镜细胞化学研究 |

【摘要】 目的 观察人血液中,老化红细胞膜上花生凝集素(PNA)和刀豆凝集素(Con A)受体细胞化学反应的变化。 方法 应用金标花生凝集素(PNAg)和刀豆凝集素(Con Ag),对早期和老化红细胞膜上PNA和Con A受体进行电镜细胞化学显示,并用计算机图像分析系统对电镜照片进行定量处理。 结果 与早期红细胞相比,老化红细胞膜上PNAg颗粒和Con Ag颗粒均有明显增多。 结论 老化红细胞膜上唾液酸的丢失导致半乳糖基和甘露糖基暴露的相应增加,这种变化可能与巨噬细胞对老化红细胞的识别和吞噬机制有关。 【关键词】 红细胞;细胞老化;刀豆凝集素;花生凝集素;细胞化学 【中图分类号】 R331.1;R329.2 【文献标识码】 A 【文章编号】 0529-1356(2000)03-268

【Abstract】 Objective To observe the cytochemical changes of peanut agglutinin (PNA)receptors and concanavalin A(Con A)receptors of aged human erythrocytes. Methods Cytochemistry of Con A and PNA labelled with colloid gold (Con Ag and PNAg)on the surface of young and aged erythrocytes was observed under electron microscope,and the micrographs were processed by computer image analysis system. Results The number of Con Ag and PNAg particles of aged erythrocytes was significantly increased in comparison with that of young erythrocytes. Conclusion The desialylation of aged erythrocytes may result in increasing exposure of galactose and mannose,which may lead to the aged erythrocytes phagocytosed by macrophages. 【Key words】 Erythrocytes;Aging;Concanavalin A;Peanut agglutinin;Cytochemistry
随着红细胞老化,膜上糖蛋白所含半乳糖和甘露糖呈下降趋势。然而,文献报道,机体内巨噬细胞和某些抗体能够识别处理老化红细胞或使这些细胞凝集,其机理可能与老化红细胞膜上半乳糖基和甘露糖基的增加有关[1]。本实验应用PNAg和Con Ag细胞化学的方法,对人早期红细胞和老化红细胞膜上半乳糖基和甘露醇糖基进行电镜细胞化学定量观察,旨在为这一问题的探讨提供依据。
材料和方法
