雌核发育银鲫与两性生殖彩鲫卵壳蛋白组分的比较分析及其受精前后的变化 |

摘 要:以雌核发育银鲫和两性生殖彩鲫的成熟卵为材料,分离卵壳,经处理得到卵壳可溶性蛋白组分。SDS-PAGE梯度凝胶电泳分析在雌核发育银鲫中揭示出3条较明显的差异蛋白带。同时,采用相同处理方法对受精前后卵壳蛋白组分进行比较分析后发现,这些差异蛋白带在受精后发生了变化,其带纹表现为减弱或消失,表明这些差异蛋白可能与受精过程相关。 关键词:雌核发育;卵壳蛋白;受精;银鲫 中图分类号:Q344 文献标识码:A 文章编号:0253-9772(2000)04-0221-04
Comparative Analysis on the Protein Composition of Egg Chorions between Gynogenetic Silver Crucian Carp and Gonochoristic Color Crucian Carp and the Changes during Fertilization

Abstracts:Egg chorions were isolated from unfertilized and fertilized eggs of gynogenetic silver crucian carp and gonochoristic color crucian carp by homogenization and further purification techniques.Then,soluble proteins were extracted from the isolated egg chorions,and were analyzed by gradient SDSPAGE.Three differential protein bands were revealed between the gynogenetic silver crucian carp and gonochoristic color crucian carp.Furthermore,these differential proteins were demonstrated to undergo obvious changes during fertilization.It was suggested that these differential proteins should be related to the special fertilization process in gynogenetic silver crucian carp. 成熟鱼类卵子表面由一层坚韧的、非细胞膜状结构所包被,这层结构即是卵壳。它在鱼类受精和胚胎发育过程中具有相当重要作用。硬骨鱼类卵壳在其动物极存在有特殊的敞开结构即受精孔(micropyle),是精子到达卵子的必经途径[1,2]。尽管鱼类卵壳存在受精孔以及精子没有顶体等结构特点使其受精过程与其他动物不同,但大量实验证明在鱼类受精过程中同样存在着精卵的特异识别作用,此外在鲤鱼及剑尾鱼等鱼类的卵子表面也发现了特定的受体分子,并获得了特定基因的克隆[3]。这些结果均提示鱼类卵壳不仅是精子入卵的机械屏障,同时还直接参与了卵子受精过程,具有一定的生物学功能。 银鲫所具有的天然雌核发育生殖方式,使其成为揭示鱼类卵母细胞发育、成熟和受精机制十分难得的独特研究系统[4]。通过采用去核、卵质转移和去除卵膜等方法[5,6,7]初步证明银鲫卵子可能存在着抑制精核发育的调控体系,而且这种调控作用因精子的来源不同而有异,显示银鲫卵子对外来精子具有某种十分精细的识别机制,不但决定着不同来源精子的不同发育方式,同时也与银鲫卵子抑制精核发育的调控系统密切相关。因此以银鲫卵壳蛋白作为研究对象,从精卵识别与早期融合的角度深入研究其受精过程,不仅有助于探讨鱼类受精过程中精卵识别和早期应答机制,[1] [2] 下一页 上一个医学论文: 采用能谱法研究铅对大鼠血脑屏障的通透性 下一个医学论文: 白腹锦鸡和红腹锦鸡的遗传分化
