晚期糖化终产物对糖尿病大鼠主动脉平滑肌细胞的影响 |

【摘要】 目的 探讨血管平滑肌细胞(VSMC)在糖尿病早期的改变及其与晚期糖化终产物(AGEs)的关系。 方法 应用荧光分光光度法、免疫组织化学、计算机图像分析和透射电镜等技术,研究链脲佐菌素诱导的糖尿病(DM)大鼠血浆和腹主动脉晚期糖化终产物(AGEs)的含量以及腹主动脉VSMC的变化。 结果 从4周起,DM大鼠血浆和腹主动脉的AGEs显著增多;腹主动脉α-SM-肌动蛋白的相对面积减小,VSMC的细胞核密度增大,线粒体和粗面内质网增多。DM大鼠腹主动脉血小板源生长因子-A的免疫组织化学反应呈持续阳性。非酶糖化抑制剂氨基胍不降低血糖,但能降低血浆和组织的AGEs水平,对VSMC的改变也有减缓作用。 结论 VSMC在糖尿病早期已出现由收缩型向合成型转化的趋势,AGEs是VSMC发生改变的重要因素。 【关键词】 糖尿病;晚期糖化终产物;血管平滑肌细胞;大鼠 【中图分类号】 R587.202 【文献标识码】 A 【文章编号】 0529-1356(2000)04-327
EFFECTS OF ADVANCED GLYCATION END PRODUCTS ON AORTIC SMOOTH MUSCLE CELL OF STZ-INDUCED DIABETIC RAT

【Abstract】 Objective The changes of vascular smooth muscle cell(VSMC) in early diabetes and their relationship with advanced glycation end products(AGEs) were studied. Methods The changes of contents of AGEs in plasma and the abdominal aorta and changes of VSMC of the abdominal aorta were investigated with methods of fluorospectrophotometry, immunohistochemistry, computer image analysis and transmission electron microscopy on streptozotocin-induced diabetic rats. Results From the fourth week, in diabetic rats, the contents of AGEs in plasma and the abdominal aorta increased, the relative area of α-SM-actin in the abdominal aorta decreased, the density of nucleus of VSMC increased, and mitochondria and rough endoplasmic reticules in VSMC increased. The immunohistochemical reaction of platelet-derived growth factor-A(PDGF-A) was persistent positive in diabetic rats. The inhibitor of nonenzymatic glycosylation, aminoguanidine hydrochloride, did not decrease the level of blood glucose, but veduced plasma and tissue AGEs, and diminish the changes of VSMC at the same time.
